A description of transdermal drug delivery has been produced which is based on the physicochemical properties of the permeant. At this time transdermal delivery is limited to the administration of potent drugs. Higher doses may be accessible if penetration enhancers are incorporated into the formulation. The kinetic model shows what properties these should have and that they are a function of the physico-chemical properties of the drug. Despite their associated problems, attempts have been made to incorporate results from ortho substituents in free energy correlations. It might be necessary, for example, to predict physico-chemical properties of a compound with an ortho substituent which acts as a drug or is a putative intermediate in a proposed mechanism. Quantitation of the ortho substituent effect requires that both steric and polar effects be considered. Another simple appHcation of the PSA is in assessing drug-like properties of molecules. Today, PSA is commonly used as an extension of the rule of 5. A high PSA indicates an increased risk of bioavailability problems for a compound. However, the predictivity of a PSA model must not be overstressed. The physico-chemical properties of a compound only influence its behavior in passive diffusion through a membrane. Other effects, such as active transport, are not considered at all. 

The pharmacological activity, efficacy, and toxicity of an administered medicament may be profoundly affected by the physico-chemical properties of the drug and the drug dosage form. Thus, such parameters as solubility, particle size, diffusional characteristics, availability and rate of dissolution of the drug have been the areas emphasized in most biopharmaceutical studies. The effect of these parameters on drug absorption is the subject of this review.
