Physical barriers breaching

The two important fuels for colonocytes are glutamine and short-chain fatty acids. The oxidation of both fuels provides ATP for the cells, which is important not only to maintain digestive and absorptive functions but also to maintain membrane structure and hence the physical barrier between the lumen and the blood and peritoneal cavity. None of the physical barriers to release should be breached. 

If the physical barriers are breached, at least one should remain intact, with no threat to its integrity. 

Every single Design Basis fault sequence has a value of less than 1 x 10, usually by many orders of magnitude. Thus the design provides for an adequate deterministic safety case for Design Basis fault sequences with the expectation that not all the physical barriers to release will be breached resulting in a minimal release of radioactivity. The fault tolerance of the facility and the effectiveness of the safety measures are thus met. 

It is therefore desirable to devise strategies both to enhance the penetration of molecules, which can already breach the skin barricade passively to some extent, and also to widen the spectrum of drug molecules that can penetrate the skin at therapeutically beneficial doses. Many tactics have been utilized to help overcome the barrier function. These include chemical means (e.g., chemical penetration enhancers or entrapment of molecules within lipid vesicles) or physical methods (such as ultrasound, microneedles, or electrical methods). Two important electrical methods are iontophoresis and electroporation. 

Strategies to breach physical and enzymatic barriers in the intestinal epithelium that hinder passage to the systemic circulation have included enhancing para-cellular flux by disrupting tight junctions. " Inhibition of the P-glycoprotein (PGP) that ejects unrecognized or unwanted materials also has been studied.f Certain lipids are reported to be PGP inhibitors but there are no reports of successful application to commercial products or use in clinical trials. 

The final group of active enhancement methods entails the use of a physical or mechanical means to breach or bypass the SC barrier. 

Detection of security breaches is usually visual, but cameras can be used to cover more ground and provide centralized coordination. Screenings also serve as detection strategies. Delay is achieved from physically securing equipment and assets, fencing, locked doors, and the systanatic use of barriers. Response at a school event may not be from law enforcement alone. Emergency medical service personnel, security officers, and trained school onployees can respond and coordinate incidents until more skilled or ranking personnel arrive (Philpott, 2010). Events should always culminate with the production of countermeasures to any incurred problems. 

Note that barrier analysis is a key step, because it fundamentally includes DiD, such as the those invoked against radioactivity release (fuel/primary system/contain-ment), and is an adaptation of the bow-tie methodology that was commonly utilized in and by the oil and gas industry. However, it is now known that such physical, procedural, administrative, and managerial layers may be breached, bypassed, or made ineffective or aggravated by human actions and subsequent loss of control, as exemplified by multiple SAs, such as the Three Mile Island loss of coolant, Davis-Besse head corrosion, Piikushima core melts and explosions, the Concorde and Air France AF447 aircraft crash, and the Deepwater Horizon offshore oil-spill events (Duffey, 2015).
