In a detailed rotating-disk electrode study of the characteristic currents were found to be under mixed control, showing kinetic as well as diffusional limitations [Ha3]. While for low HF concentrations (<1 M) kinetic limitations dominate, the regime of high HF concentrations (> 1 M) the currents become mainly diffusion controlled. However, none of the relevant currents (J1 to J4) obeys the Levich equation for any values of cF and pH studied [Etl, Ha3]. Because of the inability to quantitatively separate the two species in solution, a theoretically calculated fractionation factor of about 1.0194 at 25°C has been widely used for pn estimates (Kakihana et al. 1977). As recently shown by Zeebe (2005) and Klochko et al. (2006) the equilibrium fractionation factor appears to be significantly larger than the theoretical value of Kakihana et al. (1977) used in paleo-pH studies. Klochko et al. (2006), for instance, reported a fractionation factor of 1.0272. [Pg.46]

Alberty analyzed the anion effect on pH-rate data. He first considered a one-substrate, one-product enzyme-catalyzed reaction in which all binding interactions were rapid equilibrium phenomena. He obtained rate expressions for effects on F ax and thereby demonstrating how an anion might alter a pH-rate profile. He also considered how anions may act as competitive inhibitors. Investigators have often used pH studies to aid in the identification of active-site residues. However, care must be exercised in this regard since observed pK values may refer to macroscopic parameters rather than microscopic values and pi values of groups can be greatly affected by the immediate environment. In Figure 2 the ir-A and AV-A plots for SODS on O.OIM NaCl sub-solutions having different pH values are shown. In all cases, phase transitions from liquid-expanded to liquid-condensed state are evident ( ). Acidification of the subsolution Increases the transition pressure but the transition is less pronounced at the lowest pH studied. This is also accompanied by an expansion of the condensed part of the curve. Small negative surface potentials are observed over most areas. The highest potential is obtained for film spread on the pH 2.2 subsolution. For small areas, the surface potential attains a positive value. This may be related to reorientation of the dipole moments of the molecules which occur once a threshold surface concentration is exceeded (O. Mlnglns and Pethlca (7) studied the monolayer properties of SODS on various sodium chloride solutions (0.1, 0.01 and O.OOIM) at 9.5 C, and they showed that the monolayer is only stable on the more concentrated salt solutions (0.1 and O.OIM). Figure 8 shows a plot of the concentration of methanol produced by the hydrolysis of SiQAC at pH 4.07 in water and the nonlinear regression curve of equation (18) assuming three consecutive, irreversible first-order reactions. A summary of the observed rate constants at each pH studied is shown in Table 4. Regression fits produced R2 values of better than 0.99 for all the pH values investigated. The net charge on albumin appears to be more significant than the nature of the substrate when considering how much protein initially binds to the aqueous/solid interface. More protein adsorbed onto copper, nickel, and germanium substrates at pH 4.8, where albumin has no net surface charge, than at pH 4.0 or pH 7.4. Since no charge effects exist between the macromolecules adsorbed on the surface, high protein densities at the aqueous/solid interface would be expected.
