Pethidine hydrochloride injection

Pethidine Hydrochloride Pethidine hydrochloride solutions are not compatible with barbiturates, aminophylline, morphine sulfate, sulfadiazine sodium, methicilline sodium, nitrofurantoin, phenytoin sodium, heparin sodium, sodium iodide, diethanolamine, sulfafurazol, acyclovir sodium, liposomal doxorubicin hydrochloride, frusemide, imipenem, and idarubicin. Visual incompatibilities are reported with mixtures of pethidine hydrochloride and cefoperazone sodium or mezlocillin sodium, nafcillin sodium and minocycline hydrochloride or tetracycline hydrochloride in 5% glucose injection.82 83... 

Dose. Usually 50 to 100 mg of pethidine hydrochloride, every 4 hours, by mouth or by intramuscular injection.

Injection of Pethidine Hydrochloride, B.P, A sterile solution of pethidine hydrochloride in water for injection. 

Dilute a volume of the injection equivalent to about 0 15 g of pethidine hydrochloride with 40 ml of water in a separator and add 1 ml of 20 per cent sodium hydroxide solution. Complete as above by the B.P. method but titrating with 0 02N perchloric acid. 1 ml 0-02N = 0 005676 g Ci5H2i02N,HC1. 

Brealey and Proctor showed that chlorocresol could be determined in Injection of Pethidine Hydrochloride, J5.P., by measurement of the extinction of a diluted sample at 279 m//, the wavelength of maximum absorption of chlorocresol. Pethidine has zero absorption at this wavelength and does not interfere. 

Tilidine is structurally DL-ethyl-rranj-2-dimethylamine-1 -phenyl-cyclohex-3-ene-1 -carboxylate hydrochloride. In pharmacological studies by Herrmann et al. (68, 69) it was shown to be a strong analgesic but one which in sufficient doses tended to stimulate respiration rather than to depress it. The acute toxicity was a half to one third that of pethidine. In man it has been shown to have a clear central analgesic effect the doses recommended by the manufacturer are up to 200 mg daily by mouth or 400 mg by i.m. or i.v. injection.
