Perphenazine

The residue is caused to crystaiiize from ether/petroleum ether, and recrystailized from acetone/petroieum ether. 6.0 grams (88% of the theory) of 6-(4-methyi-1-piperazinyl)-morphanthridine of meiting point 138°-138.5°C are obtained.

A stirred mixture of 5 parts of 2-chloro-10-[7-(N-piperazino)propyl] phenothiazine, 1.92 parts of 2-bromoethanol, 2.11 parts of potassium carbonate and 35 parts of toluene is refluxed for 5 hours. The mixture is treated with water and benzene and the organic layer is separated, washed with water, dried over anhydrous potassium carbonate, filtered and evaporated. The residue is distilled at about 240°-244°C and 0.15 mm pressure to yield 2-chloro-10-[7-(N -/3-hydroxyethyl-N-piperazino)-propyl] phenothiazine according to U.S. Patent 2,838,507. 

The 2-chloro-10-( y-chloropropyl)phenothiazine starting material is produced from 2-chloro-phenothiazine and 1 -bromo-3-chloropropane.

Appropriate modification of the last few steps affords clo-penthixol (37). It is of note that this compound approaches the potency of one of the most active phenothiazines, perphenazine, an agent that has a very similar side chain.

Oral liquid concentrates are available for use in patients who can more easily swallow a liquid. These concentrates are light sensitive and dispensed in amber or opaque bottles to help protect the concentrate from light. They are administered mixed in liquids such as fruit juices, tomato juice, milk, or carbonated beverages. Semisolid foods, such as soups or puddingy, may also be used. Perphenazine (Trilafon) concentrate should not be mixed with beverages containing caffeine (coffee, cola), tea, or apple juice because of the risk of incompatibility.

Immediately after dipping perphenazine (h/ f 28-33) appeared as pink-colored, fluphenazine (hRj 30-35), dixyrazine (hRj 33-38) and verophene (= promazine, hRf 38-43) as skin-colored and periciazine (h/ f 35-40) as orange, promethazine (h/ f 43-48) as bright pink, alimemazine (h/ f 60-65) as skin-colored and dibutil (etho-propazine, hRf 75-80) as pink-colored chromatogram zones on a colorless background (Fig. 2). The colors faded after some time.
