Periodate treatment

Periodate oxidation followed by reduction and acid hydrolysis gave rise to glycerol and Xyl in a molar ratio of 88 12. However, when the polyol was reoxidized with periodate and reduced, acid hydrolysis gave rise to glycerol and Xyl in a molar ratio of 94 6, as expected. Interunit hemiacetal formation must have taken place during the initial periodate treatment inhibiting further oxidation. [Pg.554]

In some instances, reducing sugars are present that can be reductively aminated without prior periodate treatment. A reducing end of a monosaccharide, a disaccharide, or a polysaccharide chain may be coupled to a diamine by reductive amination to yield an aminoalkyl derivative bound by a secondary amine linkage (Figure 1.96). Also see Section 4.6, this chapter, for an extensive discussion on carbohydrate modification techniques. [Pg.123]

The use of periodate as a cleavage agent does have advantages, however. Unlike the use of cleavable crosslinkers that contain disulfide bonds which require a reductant to break the conjugate, cleavage of diol-containing crosslinks with periodate typically preserves the indigenous disulfide bonds and tertiary structure of proteins and other molecules. As a result, with most proteins bioactivity usually remains unaffected after mild periodate treatment. [Pg.393]

For the TBA assay, periodate treatment was conducted in the presence of 125 mM sulfuric acid (method A), or at pH 5 (method B).26 The data are given in percent, and as molar ratios (in parentheses). [Pg.344]

On studying the KDO content of LPS from Xanthomonas sinensis, Volk and coworkers64 found that only one KDO residue per LPS chain is present in this organism. Moreover, the authors observed the formation of 4,7- and 4,8-anhydro derivatives (39 and 40) of KDO (see Scheme 14) under the conditions used for mild hydrolysis of the LPS with acetic acid. Free KDO was also shown to yield the anhydro derivatives under analogous conditions. The structures of these unusual side-products were identified by mass spectrometry, as follows. Reduction of 39 and 40 with sodium borohydride gave the two epimeric mixtu.es, 41 and 42. These were subjected to periodate treatment followed by borohydride reduction, the products separated by paper electrophoresis, and their trimethylsilyl ether derivatives analyzed by... [Pg.348]

C]gulono- 1,4-lactone. The hydroxyl groups at C-2 and -3 were protected by isopropylidenation, and the 5,6-glycol was oxidized by sodium periodate. Treatment of the resulting syrupy product with methanolic hydrogen chloride, followed by borohydride reduction and hydrolysis, afforded L-[5-,4C]arabinose. [Pg.162]

However, as quantum chemical methods treat all contributions of the re-body interaction simultaneously, a periodic treatment cannot be invoked when localized basis sets are utilized and a special treatment has to be employed to maintain periodicity (27,34). Ignoring the incompatibility of the periodic environment and the re-body treatment of quantum chemical methods would lead to severe artifacts, which should be avoided at all costs. [Pg.153]

The enzymes commonly used as labels in ELISA and other immunochemical reactions include horse radish peroxidase (HRP) and alkaline phosphatase (AP). The enzyme can be covalently coupled to the antibody using glutaraldehyde conjugation to reactive amino groups on the enzyme (lysines) in a phosphate buffered aqueous solution at neutral pH, as shown in Fig. 19 (103). Alternatively, carbohydrates present in the immunoglobulin structure can be cleaved by periodate treatment (see Fig. 20) and bound to free amino groups on the enzyme through a Schiff base reaction (103). [Pg.395]

Treatment of MFA (1) with cyanogen bromide [6] opened ring G to yield the bromo derivative 3 [7]. Attempts to dehydrobrominate 3 in one step via a base-catalyzed elimination with DBU/CH3CN, KOH/MeOH, or terr-BuOK/DMSO were unsuccessful. However, the required methylene entity could be introduced by converting 3 first to a selenide, then oxidation with periodate, followed by thermolysis in benzene to provide compound 4. Hydrolysis of the cyano group with NaOH in ethylene glycol [8] produced 5 (50% yield). Osmium catalyzed oxidation of 5 in the presence of 4-methylmorpholine A-oxide (NMO) gave a diol, which was cleaved to an aldehyde upon treatment with periodate. Treatment of the aldehyde with sodium cyanoborohydride resulted in an intramolecular reductive animation to yield the desired product PHB (6). [Pg.334]

At times an induction period is observed before bromine trifluoride begins to react with the substrate Adding bromine to the substrate often shortens this induction period Treatment of 2-methoxy-l,l,l,3,3,3-hexachloropropane or 2 chloromethoxy-l,l,l,3,3,3-hexachloropropane with bromine trifluoride gives a variety of products and presents a very interesting picture of the selectivity of bromine trifluoride with and without internally generated hydrogen fluoride [S] (equations 5 and 6)... [Pg.174]

It was found that when grafting methyl methacrylate or styrene to rubber, the branches of polymethyl methacrylate or polystyrene are much smaller than was expected, while almost 50 per cent more vinyl polymer was bound than predicted (8, 27, 129, 130, 160, 161). The length of the side branches was determined after oxidation of the rubber, usually by ozonolysis (28, 71, 131) or by perbenzoic acid oxidation followed by periodate treatment (168). [Pg.187]

The lower-melting dimethylene compound was resistant to periodate treatment and its ditosyl ester did not participate in the sodium iodide exchange reaction thus it was probable that the diacetal was of the 1,2,4,6-, 1,3,4,6- or 1,3,5,6-type. For theoretical reasons the authors48 favored tentatively the 1,3 4,6-pattern. Since the other dimethylene-D-talitol (m. p. 261-262°) did not react with periodate and since its ditosyl ester could be converted into a didesoxy-dimethylene-hexitol by sodium iodide exchange and hydrogenation, it was designated as 2,3,4,5-dimethyl-... [Pg.176]

Embedding techniques at various levels have been suggested to close the gap between the cluster and the periodic treatment. A physical approach to the electronic structure problems of solids and surfaces contrasts sharply with the intuition of chemists that local interactions dominate the properties of surfaces and adsorption complexes. Hence, it is very desirable to replace the infinite solid, which is very difficult to treat quantum-chemically, by finite models of the sites of interest. In this way it is easy to describe a local site as cluster having a relatively small number of atoms which interact with a potentially infinite number of surrounding atoms through, for example, the Madelung potential which is treated as perturbation.26,44 These lead to terms such as... [Pg.572]
