Performance-on-Line

The Pascal code was updated to handle four channels. To follow the inspection speed all raw measuring data were captured and stored in the computer. All data for one coil could be stored in the computer memory (RAM) and transferred to the disk before inspection of the next coil. Evaluation of the data could be performed on-line or later using a special evaluation program. 

Blood components are also collected through apheresis. In apheresis, advanced blood cell separators are used to collect one or more specific blood components from a donor. The cell separators collect blood iato a separation chamber, isolate the desired blood components, and return the blood components not needed to the donor. This procedure is performed on-line within one sterile disposable tubiag set. The two principal components collected through apheresis are plasma and siagle-donor platelets (SDP). 

The asymptotic observer was tested in an experimental 1 m upflow anaerobic fixed bed reactor pilot plant used for the treatment of industrial wine distillery vinasses obtained from local distilleries in the Narbonne area (France) (see Figure 26). These experimental runs were carried out over a 35 day period. Measurements of the dilution rate as well as the and S2 concentrations and the partial CO2 pressure were performed on-line (see Figures 27 to 30). TG-FT-IR, Pyrolysis analyses were performed on the preliquefaction solids using thermogravimetric (TG) analysis with on-line analysis of the evolved products (including an infrared spectrum of the condensables) by FT-IR. The TG-FTIR method has been described previously (23-25). The Bomem TG/plus instrument was employed. A sample is continuously weighed while it is heated. A flow of helium sweeps the products into a multi-pass cell for FT-IR analysis. Quantitative analysis of up to 20 gas species is performed on line. Quantitation of the tar species is performed by comparison with the balance reading. 

The gases exiting the reactor pass through a Beckman 565 infrared CO2 analyzer, which continuously monitored the production of carbon dioxide. Gas composition analysis was performed on-line using a Hewlett Packard 5890 II gas chromatograph, equipped with both a thermal conductivity and a flame ionization detector and a Porapak-Q column. Additional experimental details are given elsewhere (9).

Capillary-in-capillary mixers were used for electrospray ionization mass spectrometry (ESI-MS), which allows one to perform on-line kinetic studies for a wide range of applications in chemistry, bioorganic chemistry, isotope exchange experiments and enzymology, just to name a few [133], ESI-MS is a method alternative to the traditionally employed quench-flow techniques with off-line analysis. 

Sample preparation in CE is similar to sample preparation in HPLC (Section 3.8). It is usually performed off-line in CE, because commercial instrumentation does not allow for on-line sample preparation. The exception is sample preconcentration, which is readily performed on-line. Sample preparation includes any manipulation of the sample, such as weighing, filtration, dilution, or derivatization, prior to introduction into the CE system.

It is important to recognise that Kalman filters are computationally elaborate and are not really suitable unless diere is a special reason for performing on-line calculations. 

The CUSAL-HPLC couple has been combined additionally with pre- or post-column derivatization. Thus, pre-column derivatization was used for the determination of colistin A and B in feeds following USAL, the analytes were derivatized with o-phthaldialdehyde/2-mercaptoethanol and separated by HPLC for fluorimetric detection [48]. The experimental set-up used is depicted in Fig. 4.1 OA. Another application of CUSAL-HPLC is the determination of A/-methylcarbamates in soils and food [49] (see Fig. 4.10B), where the analytes were also derivatized with o-phthaldialdehyde after separation for fluorescence-based monitoring. A number of steps of the process including leaching, filtration, solid-phase extraction, liquid chromatographic separation, post-column derivatization and fluorescence detection were performed on-line, all in an automated manner.
