Peptides detection methods

Finally, RNA editing can be involved, as in the case of the amphibian bombesin-like peptides, where nucleotides in the mRNA are changed and the final protein is not a direct reflection of the sequence encoded in the gene. RNA editing is also seen in the glutamate (Ch. 15) and serotonin receptors (Ch. 13 and Ch. 15) and probably will be found elsewhere as detection methods become more sophisticated. [Pg.326]

Antibody-based detection methods include immuno-cytochemistry, which gives qualitative data but has very good spatial resolution. Radioimmunoassays provide a quantitative measure of release or content. One of the major limitations of all antibody-based methods is the potential for cross-reactivity among the many peptides. For example, some of the most sensitive gastrin antisera also detect CCK, since the peptides share a common COOH-terminal tetrapeptide sequence. Methods for detection of the mRNAs encoding neuropeptides include Northern blots, which provide quantitative data and information on splice variants, but lack fine anatomical resolution. The more commonly used polymerase chain reaction, which can be quantitative but often is used in a more qualitative manner, provides great sensitivity. Alternatively, in situ hybridization preserves anatomical relationships and can be used to obtain both qualitative and quantitative data. [Pg.328]

For analyses of synthetic peptides that are generally available in larger amounts than isolated natural peptides and proteins, the ninhydrin detection method is fully sufficient. The alternative detection method with orthophthaldialdehyde, which improves the sensitivity by a factor of five, was originally proposed for post-column derivatization, 34 but is presently used mainly in pre-column derivatization techniques. 35,36 ... [Pg.654]

Peptides complementary to the surface of Ab-2 (which acts as a receptor bioisostere) are selected from a peptide library (Lam et al., 1991 Houghten et al., 1991) using molecular immunology detection methods. Such peptides are expected to be receptor antagonists or agonists (Scott and Smith, 1990). [Pg.116]

These results validated the ELISA detection method for stem peptide. In addition, the other components of the enzyme reaction mixture were tested in the ELISA, and none cross-reacted with the antibody at concentrations used in the assay. [Pg.299]

In this chapter, we will present different types of biochips, including protein and antibody arrays, as well as carbohydrate, peptide and living cell arrays. We will discuss recent progress and current bottlenecks in high-throughput generation of chip content, surface chemistry, molecule attachment, detection methods, and also applications in the proteomic field and in drug discovery. [Pg.139]
