Peptide station

An important feature of the rotaxane in Scheme 9.12 is that the translocation of the macrocycle is also achieved by the reduction of the fullerene to its trianion, which is both effected and observed by cyclic voltammetry. In DMSO, the proximity of the macrocycle to the fullerene stabilized substantially the electrogenerated trianion (A i/2 = 46 mV) through n—n interactions. Surprisingly, in THF where the macrocycle is preferentially positioned on the peptide station, a similar behavior was... [Pg.241]

Vesicular proteins and lipids that are destined for the plasma membrane leave the TGN sorting station continuously. Incorporation into the plasma membrane is typically targeted to a particular membrane domain (dendrite, axon, presynaptic, postsynaptic membrane, etc.) but may or may not be triggered by extracellular stimuli. Exocytosis is the eukaryotic cellular process defined as the fusion of the vesicular membrane with the plasma membrane, leading to continuity between the intravesicular space and the extracellular space. Exocytosis carries out two main functions it provides membrane proteins and lipids from the vesicle membrane to the plasma membrane and releases the soluble contents of the lumen (proteins, peptides, etc.) to the extracellular milieu. Historically, exocytosis has been subdivided into constitutive and regulated (Fig. 9-6), where release of classical neurotransmitters at the synaptic terminal is a special case of regulated secretion [54]. [Pg.151]

Early reported applications of this technique were the preparation of a 24-member peptide library [83], of a 125-member tripeptide-substituted cinnamic acid library tested for inhibition of tyrosine phosphatase PTP1B [83], of a 64-member peptide-like library [83] and of libraries based on a natural product, epothilone, using also new polystyrene grafted solid supports [84], Other applications, ranging from l,5-benzodiazepin-2-one library synthesis [85] to chalcone library synthesis [86], were also recently reported. Commercialization of the basic components for this technique [87] (reaction supports and vessels, tags, software, sorters, reaction stations, and so on) will ensure its quick and effective use in combinatorial chemistry and also the implementation of new technical features and possibilities for more complex and demanding applications in future. [Pg.225]

CEM also offers a manual peptide synthesizer. Discover SPS (Fig. 20.7). This system is also based on the Discover microwave reactor. It uses a 25 mL polypropylene SPE cartridge that is held in place in the reactor. Fiber-optic temperature monitoring is included in the system and enables control of microwave power delivery. A vacuum manifold station is provided that enables rapid washing and filtration of reagents during the cycle. The system can be upgraded to the automated Liberty. [Pg.907]
