The surprising discovery that small oligo-P-peptides exhibit extraordinary tendencies to form stable secondary structures has led to rapid developments in the chemistry of these peptides. The earliest work in the field revolved around the synthesis of P-peptide polymers (the so-called nylon-3 derivatives). 2 Polymerization of P-amino acids led to polymers of undefined length. It was noted they could form stable structures but it proved impossible to gain any concrete information about the nature of those structures at that time. The developments in the synthesis of P-peptide oligomers of predefined length and advances in methods (i.e., NMR spectroscopy and X-ray crystallography) for the 3D characterization of such compounds has led to the discovery of new helical structures found to be adopted by a variety of P-peptides. I1,3-7 ... [Pg.552]

The synthesis of peptidyl polymers has a long and elegant history, beginning with the construction of homopolypeptides and random copolypeptides. More recently, biomolecules that incorporate peptide entities into classic polymer structures have been created. Sequential peptide polymers have been constructed that model the three-dimensional structure of connective tissue proteins. This article describes methods for the assembly of a variety of peptidyl polymers. [Pg.169]

Approaches to artificial ion channels have, for instance, made use of macrocyclic units [6.72,6.74] (see also below), of peptide [8.183-8.185] and cyclic peptide [8.186] components, of non-peptidic polymers [8.187] and of various amphiphilic molecules [6.11, 8.188, 8.189]. The properties of such molecules incorporated in bilayer membranes may be studied by techniques such as ion conductance [6.69], patch-clamp [8.190] or NMR [8.191, 8.192] measurements. However, the nature of the superstructure formed and the mechanism of ion passage (carrier, channel, pore, defect) are difficult to determine and often remain a matter of conjecture. [Pg.114]

Borner and coworkers demonstrated that peptide-polymer conjugates can assemble to nanotapes with persistence lengths close to those of actin filaments (actin composes one part of the intercellular skeleton) [108, 109]. The generated nanostructures are flat ribbons (1.2 nm x 17 nm x 2 pm height x width x length) with... [Pg.172]
