Tap Density. Tapping a mass of loose powder, or more specifically, the appHcation of vibration to the powder mass, separates the powder particles intermittently, and thus overcomes friction. This short-time lowering of friction results in an improved powder packing between particles and in a higher apparent density of the powder mass. Tap density is always higher than apparent density. The amount of increase from apparent to tap density depends mainly on particle size and shape (see Table 4). [Pg.181]

Investigation of the differences in crystal packing between (431) and (426) from comparison of their respective X-ray structures, revealed that (431) was more tightly packed than (442), reflected in their respective melting points of 235 and 170 °C. It was postulated that the absence of in vivo activity for (431) may be explained by the resultant reduction in water solubility and dissolution rate compared with (426). The comparatively high calculated polar surface area of (431) (122.5A ) compared with (426) (89.3 A ) was also proposed as a factor influencing the marked difference in bioavailability between the two related compounds. Compound (426) (SLV-319) is currently being developed with Bristol-Myers Squibb for the potential treatment of obesity and other metabolic disorders. Phase I trials for obesity were started in April 2004. Earlier Phase I clinical trials for the treatment of schizophrenia and psychosis, which commenced in April 2002, appear to have been abandoned. [Pg.285]

Photolyses were performed at room temperature using a Nagano Science LT-120 irradiatior equipped with a Toshiba chemical lamp FIR-20S-BL/M (800 mW cm-2). Initially, we attempted to build a 3-D homology model of the transmembrane regions of the human thrombin receptor from the structure of IBRD, which is available from the Brookhaven Protein Database. Unfortunately, the suboptimal placement of several amino acid side chains resulted in severe deviations from structural standards for membrane-bound receptors with a seven-helix bundle topology (Figure 2). For example, carboxylate and ammonium groups on amino acid side chains at a mid-helix location were directed into the membrane, rather than toward the inside of the helix bundle, and the hydrophobic packing between some helices was either poor or nonexistent. [Pg.250]
