Finally, the recording of many signals from the output of the analytic and electrochemical instrumentation requires a reliable multi-pen recorder or an equivalent recording system based on a data acquisition card and appropriate software. Generally, software sensors are typical solutions of so-called inverse problems. A so-called forward problem is one in which the parameters and starting conditions of a system, and the kinetic or other equations which govern its behavior, are known. In a complex biological system, in particular, the things which are normally easiest to measure are the variables, not the parameters. In the case of metabolism, the usual parameters of interest are the enzymatic rate and affinity constants, which are difficult to measure accurately in vitro and virtually impossible in vivo [93,118,275,384]. Yet to describe, understand, and simulate the system of interest we need knowledge of the parameters. In other words, one must go backwards from variables such as fluxes and metabolite concentrations, which are relatively easy to measure, to the parameters. Such problems, in which the inputs are the variables and the outputs the parameters, are known as system identification problems or as so-called inverse problems. [Pg.36]

A posteriori identifiability is linked to the theory of optimization in mathematics because one normally uses a software package that has an optimization (data-fitting) capability in order to estimate parameter values for a multicompartmental model from a set of pharmacokinetic data. One obtains an estimate for the parameter values, an estimate for their errors, and a value for the correlation (or covariance) matrix. The details of optimization and how to deal with the output from an optimization routine are beyond the scope of this chapter, and the interested reader is referred to Cobelli et al. (12). The point to be made here is that the output from these routines is crucial in assessing the goodness-of-fit — that is, how well the model performs when compared to the data — since inferences about a drug s pharmacokinetics will be made from these parameter values. [Pg.102]

One problem arises in DAD use that was not as major in single-wavelength detection, the absorbance of the mobile phase. In single-wavelength detection under isocratic (constant composition) conditions, any absorbance due to the mobile-phase solvents only resulted in a constant increase in the baseline. This was easily overcome by normalizing the basehne output to zero. If gradient elution was used, the baseline rose in a continuous, readily accounted for fashion. With DADs, the total absorbance of the mobile-phase interferes with spectral collection in that range. Deconvolution software, however, can extract the spectra. [Pg.988]

Most integrators perform area percent, height percent, internal standard, external standard, and normalization calculations. For nonlinear detectors, multiple standards can be injected, covering the peak area of interest, and software can perform a multilevel calibration.
