Nitroxides reaction with radicals

Stable radicals can show selectivity for particular radicals. For example, nitroxides do not trap oxygcn-ccntcrcd radicals yet react with carbon-centered radicals by coupling at or near diffusion controlled rates.This capability was utilized by Rizzardo and Solomon " to develop a technique for characterizing radical reactions and has been extensively used in the examination of initiation of radical polymerization (Section 3.5.2.4). In contrast DPPH, while an efficient inhibitor, shows little selectivity and its reaction with radicals is complex. ... 

Adding a radical trap like BulNO to the reaction mixture this reacts with radicals (R ) forming nitroxide radicals Bul(R )NO that can be detected by ESR. 

Most radicals are transient species. They (e.%. 1-10) decay by self-reaction with rates at or close to the diffusion-controlled limit (Section 1.4). This situation also pertains in conventional radical polymerization. Certain radicals, however, have thermodynamic stability, kinetic stability (persistence) or both that is conferred by appropriate substitution. Some well-known examples of stable radicals are diphenylpicrylhydrazyl (DPPH), nitroxides such as 2,2,6,6-tetramethylpiperidin-A -oxyl (TEMPO), triphenylniethyl radical (13) and galvinoxyl (14). Some examples of carbon-centered radicals which are persistent but which do not have intrinsic thermodynamic stability are shown in Section 1.4.3.2. These radicals (DPPH, TEMPO, 13, 14) are comparatively stable in isolation as solids or in solution and either do not react or react very slowly with compounds usually thought of as substrates for radical reactions. They may, nonetheless, react with less stable radicals at close to diffusion controlled rates. In polymer synthesis these species find use as inhibitors (to stabilize monomers against polymerization or to quench radical reactions - Section 5,3.1) and as reversible termination agents (in living radical polymerization - Section 9.3). 

Nitrones arc generally more stable than nitroso-compounds and arc therefore easier to handle. However, the nitroxides formed by reaction with nitrones [e.g. phenyl /-butyl nitrone (109)]483 484 have the radical center one carbon removed from the trapped radical (Scheme 3.86). The LPR spectra are therefore less sensitive to the nature of that radical and there is greater difficulty in resolving and assigning signals. Nitrones are generally less efficient traps than nitroso-compounds.476... 

Some limitations of the method arise due to side reactions involving the nitroxide. However, such problems can usually be avoided by the correct choice of nitroxide and reaction conditions. Nitroxides, while stable in the presence of most monomers, may act as oxidants or rcductants under suitable reaction conditions.516 The induced decomposition of certain initiators (e.g. diacyl peroxides) can be a problem (Scheme 3.94).166 177 There is some evidence that nitroxides may disproportionate with alkoxy radicals bearing a-hydrogens,123 Side reactions with thiols have also been identified.4 18... 

The reaction of radicals with nitroxides is reversible. 09 This means that the highest temperature that the technique can reasonably be employed at is ca 80 °C for tertiary propagating species and ca 120 °C for secondary propagating species.22 These maximum temperatures are only guidelines. The stability of alkoxyamines is also dependent on solvent (polar solvents favor decomposition) and the structure of the trapped species. This chemistry has led to certain alkoxyamines being useful as initiators of living polymerization (Section 9.3.6). At elevated temperatures nitroxides are observed to add to monomer albeit slowly. 3IS 5" 523... 

The efficiency of these inhibitors may depend on reaction conditions. For example the reaction of radicals with stable radicals (e.g. nitroxides) may be reversible at elevated temperatures (Section 7.5.3) triphenylmethyl may initiate polymerizations (Section 7.5.2). A further complication is that the products may be capable of undergoing further radical chemistry. In the case of DPPH (22) this is attributed to the fact that the product is an aromatic nitro-compound (Section 5.3.7). Certain adducts may undergo induced decomposition to form a stable radical which can then scavenge further. 

Aromatic nitro-compounds have also seen use as inhibitors in polymerization and as additives in radical reactions. The reactions of these compounds with radicals are very complex and may involve nitroso-compounds and nitroxide intermediates.20" 206 In this case, up to four moles of radicals may be consumed per mole of nitro-compound. While nitroxides give overwhelmingly combination in their reaction with carbon-centered radicals, the amount of disproportionation is finite (Scheme 9.24). Disproportionation cannot always be rigorously distinguished from elimination and it is possible that both reactions occur. The combinatiomdisproportionation ratio (or extent of elimination) depends on the nitroxide and radical structure and within a scries of structurally related systems appears to increase as... 

In a study directed to the analysis of the role of Fe and the generation of H2O2 in Escherichia coli (McCormick et al. 1998), hydroxyl radicals were specihcally trapped by reaction with ethanol to give the a-hydroxyethyl radical. This formed a stable adduct with a-(4-pyridyl-l-oxide)-iV-t-butyl nitroxide that was not formed either by superoxide or hydroxyl radicals. The role of redox-reactive iron is to use EPR to analyze the EPR-detectable ascorbyl radicals. 

The reactions of radicals with oxygen are diffusion-controlled it-S-S. Moreover, as has been previously shown, the isobutyryl radical a could readily be captured by a nitroxide. It is therefore not easy to see why reaction between oxygen and the species a does not also occur. 

If one takes into account not only the initial slope of the curves but also the part played by the formation of isobutyrate it can be seen that the amount of reaction products formed is almost equivalent to the loss of DiPK. In this case the formation of isobutyric acid represents the most important difference compared with irradiation without additive. It shows that in the presence of nitroxide the acyl radical may not only be captured by oxygen but can also react further as acyl-peroxy radical, without losing its carbonyl group in the process. 

A second mechanism involving as intermediate step a stable hydroxylamine ether (isopropyl I-ether) is also a possibility (reaction (15)). In a second step the ether would undergo cleavage by the acylperoxy radical with formation of isobutyric acid and acetone and liberation of the nitroxide (reaction (16)) ... 

N-AryInitrones (XIII) formed by oxidation of N-hydroxy-N-methyl arylamines, show high reactivity toward carbon-carbon and carbon-nitrogen double bonds in non-aqueous media (21,203) (Figure 10). Under physiological conditions, however, it appears that N-arylnitrones exist as protonated salts that readily hydrolyze to formaldehyde and a primary N-hydroxy arylamine and efforts to detect N-arylnitrone addition products in cellular lipid, protein or nucleic acids have not been successful (204). Nitroxide radicals derived from N-hydroxy-MAB have also been suggested as reactive intermediates (150), but their direct covalent reaction with nucleic acids has been excluded (21). 

Unfortunately, due to the above shortcomings of hydroxylamine derivatives as spin traps, the uncertainties of the mechanism of their reactions with superoxide are added. Although it is supposed that nitroxide radicals are formed by oxidation with superoxide (Reaction (6)), this reaction cannot be an elemental stage because superoxide cannot abstract a hydrogen atom. 

Decomposition of azo compounds and peroxides provides the alkoxyamine by the nitroxide-trapping of the primary radicals [29]. The radicals produced by hydrogen abstraction with oxy radicals are also trapped by the nitroxide [242, 243]. In the photoreaction, alkoxyamines were isolated with high yields [244]. The reactions of Grignard reagents with nitroxides [215] and the coupling reaction of sodium nitroxides with bromo compounds [234,235] are also used. The hydrolysis of 56 followed by the reaction with acyl or alkyl halides afforded alkoxyamines with various functional groups,63 (Eq. 68) [245-251] ... 

If di(tcrt-butyl)nitroxide (a radical trap) is present, the reaction with phenylacetonitrile-potassium does not proceed entirely. Acetonitrile-potassium (which is in equilibrium with potassium amide) forms only aminopyridine in the presence of the trap (Moon et al. 1983). Consequently, amination is a classical nucleophile reaction, and the formation of pyridyl acetonitrile is a reaction of the typs- These two reactions are quite different. A stronger CH acid leads to a well-defined synthesis. 

The treatment of C q with an excess of bis(trifluoromethyl)nitroxide [(CF3)2NO radicals] leads to multiple additions with an average of 18 (Cp3)2NO groups bound to the fuUerene sphere (Scheme 6.16) [74]. The reaction has been carried out in a glass tube equipped with a Teflon valve. The resulting light brown solid decomposes above 158 °C. 

The photochemical transformation of the spirocyclic 4//-pyran derivative 622 shown in Eq. (47)333 resembles the reverse of the phototransformation 349->350. Upon reaction with Pb02 622 gave a stable nitroxide radical.454... 

A radical ipso substitution at the 3-position of 2,3-disubstituted indoles has also been reported in their reaction with benzoyl-r-butyl nitroxide leading to (227) or, with the 2-substituted indole, the dimer (228) (cf. Section 3.05.1.4) (81CC694). In contrast with the benzoyloxylation reactions the nitroxide radical initially abstracts the hydrogen atom at the 1-position to form the indolyl radical. This mechanism is supported by the failure of the corresponding 1 -methylindole to undergo an analogous oxidation. 

Under photo-stimulation, isoindolyloxyl radical (5) abstracts primary, secondary, or tertiary hydrogens from unactivated hydrocarbons including cyclohexane, isobutane, or n-butane (Scheme l).23 The nitroxide (5) traps the resultant carbon-centred radical (R ) and so afford the A -aI koxyisoindo les (6). Blank photolysis experiments with no added hydrocarbon have shown some unprecedented / -fragmentation of (5) to afford the nitrone (7). A number of C60 nitroxide derivatives have been synthesized and characterized by ESR spectroscopy which show features common to nitroxide radicals.24 Reaction of nitroxide and thionitroxide radicals with thiyl radicals have been observed, from which sulfinyl, sulfonyl, and sulfonyloxy radicals were generated.25 The diisopropyl nitroxide radical was generated in the reaction of lithium diisopropylamide with a-fluoroacetate esters.26... 

An investigation into the initiation mechanism of copolymerization of ethyl vinyl ether and acrylonitrile by /-butoxyl radicals lias shown that the reaction between the two monomers competes successfully with radical trapping by the nitroxide radical trap (5).37 The /-butoxyl radicals react 3-6 times faster with ethyl vinyl ether than acrylonitrile the authors proposed that this is due to selective interaction of one monomer with the radical species rather than a solvent polarity effect.
