Methadone hydrochlorid

Methadone hydrochloride (2-dimethylamino-4-ethoxycarbonyl-4,4-diphenylbutane HCl) [W95-90-5J M 345.9, m 241-242 , pKj 8.94, pK 10.12 (free base). Crystd from EtOH. 

Diphenylacetonitrile is condensed with 2dimethyl-amino)-2,2-diphenyl valeronitrile. It is then reacted with ethyl magnesium bromide and then hydrolyzed using HCl to give methadone hydrochloride.

METHADONE (METHADONE HYDROCHLORIDE) Like LAAM, a synthetic opiate used to treat heroin addiction. Methadone is non-intoxicating and blunts symptoms of withdrawal. 

Methadone hydrochloride (600.0 mg,1.74 mmol) was dissolved in ethanol (10 ml) and the solution was stirred whilst sodium borohydride (3.47 mmol) was added portion-wise over a period of 5 min. When the addition was complete a spatula end of cerium (III) chloride heptahydrate was added. The resultant solution was allowed to stir at room temperature for 30 min then the ethanol was removed under reduced pressure. The residue was portioned between diethyl ether (40 ml) and water (40 ml). The aqueous layer was extracted with more diethyl ether (2 times 20 ml) and then the combined organics were washed with brine (40 ml) and dried (MgS04). The ether was removed under reduced pressure to leave 435.0 mg, (80%) of 6-dimethylamino-4,4-diphenyl-3-heptanol. 

Sample preparation procedures for GC are generially more involved. For example, for methadone hydrochloride, 0.5jV sodium hydroxide is added to give the free base, followed by extraction with methylene chloride. An internal standard is added after the extract is dried with anhydrous sodium sulfate [3, p. 970]. The assay of interleukin-la formulated with human serum albumin does not require any sample treatment prior to analysis by capillary electrophoresis [44]. 

Methadone Hydrochloride Visual incompatibilities of methadone hydrochloride were observed with solutions of aminophylline, ammonium chloride, amylo-barbitone sodium, chlorothiazide sodium, heparin sodium, nitrofurantoin sodium, novobiocin, pentobarbitone sodium, phenobarbitone sodium, phenytoin sodium, quinalbarbitone sodium, sodium bicarbonate, sodium iodide, sulfadiazine sodium, sulfafurazole diethanolamine, or thiopentone sodium.
