Praziquantel was developed in the late 1970s (Gonnert and Andrews, 1 977) and tested in humans soon after (Katz et al., 1979 Santos et al., 1979). It has been identified by the World Health Organization (WHO) as the drug of choice for the treatment of schistosomiasis caused by S. mansoni, S. japonicum or 5. haematobium since 1982 (Andrews et al., 1983). The mechanism of praziquantel s anti-schistosomal activity remains unclear, but recent insights are reviewed in Chapter 14. More information about other uses for praziquantel for other flatworm diseases can be found in Chapter 12.

Recently insight was gained in the mechanism of oxidizing indolines to 5-hydroxyindoles with Fremy s salt B. Giethlen, J. M. Schaus, Tetrahedron Lett. 1997, 49, 8483-8486. 

QSAR Correlation. Statistical quality of QSAR correlation can be employed as a third criterion of commonality of mechanism. This approach can prove very meaningful when coupled with a mechanistic interpretation of the role of molecular descriptors used in the correlation, and with the significance of the slope and intercept. The quality of statistical fit and the interpretation of the parameter or parameters used in the correlation can provide a valuable insight into molecular mechanism. Recently, Hansch analysis has been combined with molecular graphics and modeling studies in which the activities of a series of substrates to an enzyme receptor have been related to the hydrophobic, electronic, and steric requirements for reversible binding... 

The development and optimization of chemical EOR systems can best be achieved on the basis of a good understanding of the mechanisms by which oil is displaced from a porous matrix, and of the parameters which control those mechanisms. Better insight into the displacement processes is required than is afforded by "black box" experiments such as displacement tests in sand packs. It is therefore necessary to carry out laboratory experiments concerned with specified system parameters, and to link the results of these experiments with displacement tests. The most commonly applied measurement used in this way is of interfacial tension, which is linked to displacement tests via intuitive conceptual models of the displacement process. Only relatively recently have systematic studies been undertaken (5) to test these models. 

In conclusion, although recent insights into the intestinal absorption of cholesterol have also provided new evidence regarding the potential action of PS, the molecular mechanisms of PS are still a field of debate and further research is necessary.

More recent insights suggest that there may be a useful window in altering the duty cycle when mechanical devices are used, exchanging the clinical sinusoidal waveform for a more trapezoidal one. 

A considerable number of experimental extensions have been developed in recent years. Luckliam et al [5] and Dan [ ] review examples of dynamic measurements in the SFA. Studying the visco-elastic response of surfactant films [ ] or adsorbed polymers [7, 9] promises to yield new insights into molecular mechanisms of frictional energy loss in boundary-lubricated systems [28, 70]. 

Several early interpretations of the polymerization mechanism have been proposed (1,17,29—31). Because of the complexity of this polymerization and insoluble character of the products, key intermediates have not ordinarily been isolated, nor have the products been characterized. Later work, however, on the resinification of furfural (32,33) has provided a new insight on the polymerization mechanism, particularly with respect to thermal reaction at 100—250°C in the absence of air. Based on the isolation and characterization of two intermediate products (9) and (10), stmcture (11) was proposed for the final resin. This work also explains the color produced during resinification, which always is a characteristic of the final polymer (33). The resinification chemistry is discussed in a recent review (5).
