It is also interesting to note that the big change in SAR occurred at sites of the molecule that probably are not consistent with easy library chemistry in all of these examples. A point should be emphasized that perhaps many molecules could have been made at the amide positions for 13 or 15 (perhaps the easiest chemistry to access), but the key changes occurred in the scaffold or the linker by addition of a methyl group. This is not to say that these amides should not be modified in a hit-to-lead campaign. After many years of debate and discussion, there is now an international consensus about the statistical approach and method to be employed in full collaborative trials This topic is discussed further in Section 9.5. The lUPAC protocol referred to above requires the analysis of duplicate test samples of the same material (a minimum of five materials or, exceptionally, three) in eight or more laboratories. There are two forms of the f-test, and each is applicable for sets of measurements that have been obtained in different ways.
