Leaks are, besides loss of power, the most unwanted events during freeze drying (see the end of this chapter about defects). Therefore it is recommended, to make a leak test before each freeze drying run, though this routine test does not have to follow the procedure described above. Once per day, the molecular masses that could indicate a leak should be checked. These are 18 (water), 28 (nitrogen) and 32 (oxygen). A high level of these masses indicates a leak in the system, e.g., at the interface between the GC and the MS. If a leak is not found by routine testing, it can lead to an increased background and consequently poor analytical results. In the worst case, it can result in destruction of the filament. 

Failures of pressure vessels are veiy rare. Many of those that have been reported occurred during pressure test or were cracks detected during routine examination. Major failures leading to serious leaks are hard to find. 

For gaseous sterilization procedures, elevated temperatures are monitored for each sterilization cycle by temperature probes, and routine leak tests are performed to ensure gas-tight seals. Pressure and humidity measurements are recorded. Gas concentration is measured independently of pressure rise, often by reference to weight of gas used. 

Water pressure is a key consideration in terms of the adequacy of application and system operation and maintenance. While some domestic supplies have high pressure systems, most are in need of pressurization, as is water from bodies of water and tanks. Fire pumps are routinely used, most often in multiple installations with different means of power drive (e.g., electric, diesel, or steam). When fire pumps operate at high pressures, there is the potential for leaks and line fracture. Periodic maintenance, including hydrostatic testing, will help minimize the potential for such failures.

Sterilization by moist heat is suitable only for water-wetable materials and aqueous solutions. Both temperature and pressure should be used to monitor the process. The temperature recorder should normally be independent of the controller, and there should be an independent temperature indicator, the reading from which is routinely checked against the chart recorder during the sterilization period. For sterilizers fitted with a drain at the bottom of the chamber, it may also be necessary to record the temperature at this position, throughout the sterilization period, here should be regular leak tests on the chamber when a vacuum phase is part of the cycle. 

Monitoring should be carried out routinely and include frequent leak testing of the isolator and glove/sieeve system. 

Isolators should have the facility to enable routine leak testing and particle counts to be carried out in the isolator itself and in its transfer devices. Where access points are provided for test equipment they should be labelled. 

As vacuum is pulled on the system, routine frequent tests of air leakage (i.e., oxygen analyses) must be conducted. If an excessive leak is detected and cannot be taken, the unit may require shutdown and repair. 

Protein molecules are too large to pass through cell membranes and are contained inside the normal cells where they were formed. However, if cells become damaged by disease or trauma, the protein contents can leak out. Thus, persistent excessive amounts of proteins in the urine are indicative of damaged kidney cells. A routine urinalysis usually includes a test for protein. Similarly, a heart attack can be confirmed by the presence in the blood of certain proteins (enzymes) that are normally confined to cells in heart tissue (Section 10.9). 

The OSHA Laboratory Standard does not mandate medical surveillance for all laboratory workers. The employer must provide workers an opportunity for medical attention. This includes follow-up examinations and treatment recommended by an examining physician when an employee exhibits signs or symptoms associated with exposure to a hazardous chemical or the worker is routinely exposed above the action level or PEL for a regulated substance. Offer medical consultation to any employee potentially exposed through a spill, leak, or explosion of a hazardous chemical or substance. Employers must provide information about the hazardous chemical, conditions under which the exposure occurred, and a description of symptoms experienced by the woiker. Employers must obtain from a treating physician any written opinion requiring follow-up examinations or medical tests.
