Infrared and Raman spectra of A-4-thiazoline-2-thione and of isotopi-cally labeled derivatives (56. 59) were interpretated completely. (Table VII-41. 

Matrix effects were evaluated by Gago-Ferrero et al. [23]. Both signal suppression and signal enhancement were observed. The extent of these effects was found to be fairly dependent on the UV filter. Thus, quantification should be performed by standard addition or internal standard calibration. Since standard addition is a high time-consuming procedure, internal standard calibration with appropriate isotopi-cally labeled compounds is the best option. 

The parent compound HBNH was the first iminoborane to be identified unequivocally (8). It was formed by photolysis of solid H3BNH3 and trapped, together with further products, in a rare gas matrix which provided low temperature and high dilution for this extremely unshielded molecule. Evidence for the iminoborane in the mixture came from a vibrational analysis, all four atoms in turn being isotopi-cally labeled. 

These are, without doubt, somewhat artifieial systems. For real systems, one could not tell if anything happened, unless isotopie labeling could be arranged. An even greater problem would oeeur in the gas phase, sinee the entropy penalty required for these peeuliar geometries would be expected to make them very improbable.

In the case of FBDD, structural enablement means having either suitable crystals for X-ray or a suitably isotopi-cally labeled protein sample for NMR. Obtaining suitable crystals for X-ray can take anywhere from 4 (literature precedent) to 18 (novel) months. These times presume that protein production must be developed in-house. In cases where high-quality proteins can be readily purchased, we have seen on rare occasions this time to structural enablement cut down to approximately 2 months. 

Once the appropriate conditions have been found to isolate chemistry, isotopi-cally labeled substrates can then be used to probe further for nuclear tunneling during the chemical step on the enzyme. There are three limiting kinetic relationships that affect which types of isotope effects can be used to study enzyme chemistry. The first case is when kcat is fully limited by chemistry, the second is when kcst/Ku is limited by chemistry, and the third arises when multiple steps are partially rate limiting. A very elegant experiment to show a similar effect was conceived and executed by Russell, Wool and co-workers (Russell et al. 1993).
