Intrinsically processing methods

Nonspecific protein binding to the solid phase complicates the method and is a selective pressure driving its evolution. The adaptive response has been the development of intrinsically comparative methods in which specific binding to an immobilized ligand is blocked in one out of two otherwise identical samples. When the respective protein components of the samples are compared, specifically bound proteins are present in one but severely depleted in the other. To allow relative quantitation, the two samples can be made isotopically distinct by a chemical or metabolic process and then mixed for an analytical step that avoids intersample variability [15]. [Pg.348]

Pyrochemical processing methods may offer unique advantages over more conventional aqueous methods with respect to meeting nonproliferation goals. Some pyrochemical processes are intrinsically proliferation resistant because the process is incapable of producing a weapons-usable product without significant alterations. The product also can be sufficiently radioactive that it is physically difficult to divert. Spectrophotometric procedures, including absorptimetric and fluo-rimetric, will continue to be popular and important because they can be sensitive and accurate, and because the necessary instruments are now famihar laboratory tools. Intrinsically, fluorimetric methods can be several orders of magnitude more sensitive than absorptimetric. To date, most direct fluorimetric methods are for the determination of metals, while anions are determined by their quenching action on fluorescent compounds. Hopefully, other new direct fluorimetric reagents for anions similar to the one described above for cyanide ion using quinone monoxime benzene sulfonate ester will be developed either as the result of chance observations or the application of increased fundamental knowledge of the fluorescence process. Electrochemistry of LB films of fullerenes has been widely studied and remains the subject of much research effort from both theoretical and experimental approaches. Bard etal. have studied basic electrochemistry of Ceo fullerene LB films on an electrode in acetonitrile solutions [23]. The study indicated that reduction of the fullerene films could form insoluble films with incorporated electrolyte cations or lead to dissolution. The study on Cgo LB films has become a focus of considerable interest however, it is difficult to fabricate high-quality LB films of pure Cgo due to its intrinsic hydropho-bicity. Kajiyama et al. applied a multistep creep method as an LB technique for constructing a fairly homogeneous Ceo monolayer, which is regularly packed in a hexagonal array [44]. Kunitake etal. developed the electrochemical replacement method to form epitaxial adlayers of fullerenes on Au(lll) surfaces [45]. The wet process method consists of the transfer of Langmuir films of fullerene onto iodine-modified Au(lll) surfaces at an air-water interface followed by the electrochemical removal and replacement of iodine adlayers with fullerene adlayers in solution. The most powerful organizing force in molecular assembly is certainly represented by the strong, highly selective, and directional HBs. Several techniques have been used for the HB detection and characterization, but due to their intrinsic limitation, methods of choice are mainly IR and NMR spectroscopy.
