Initiation complexes

Antibiotic A201A. Antibiotic A201A (23), produced by S. capreolus is an /V -dimethyladenine nucleoside stmcturaHy similar to puromycin (19). Compound (23) which contains an aromatic acid and monosaccharide residues (1,4), inhibits the incorporation of amino acids into proteins but has no effect on RNA or DNA synthesis. Compound (23) does not accept polypeptides as does (19), and does appear to block formation of the initiation complex of the SOS subunit. It may block formation of a puromycin-reactive ribosome. [Pg.122]

Examine the sequence of structures corresponding to Ziegler-Natta polymerization of ethene, or more specifically, one addition step starting from a zirconocene-ethene complex where R=CH3. Plot energy (vertical axis) vs. frame number (horizontal axis). Sketch Lewis structures for the initial complex, the final adduct and the transition state. Indicate weak or partial bonding by using dotted lines. [Pg.251]

The mechanism of organolithium addition to naphthyl oxazolines is believed to occur via initial complexation of the alkyllithium reagent to the oxazoline nitrogen atom and the methyl ether to form chelated intermediate 17. Addition of the alkyl group to the arena 7t-system affords azaenolate 18, which undergoes reaction with an electrophile on the opposite face of the alkyl group to provide the observed product 4. The chelating methyl... [Pg.239]

Oxazolidinones are a new class of synthetic antimicrobial agents, which have activity against many important pathogens, including methicillin-resistant Staphylococcus aureus and others. Oxazolidinones (e.g. linezolid or eperezolid) inhibit bacterial protein synthesis by inhibiting the formation of the 70S initiation complex by binding to the 50S ribosomal subunit close to the interface with the 3OS subunit. [Pg.919]

Given that these proteins have properly assembled, the initiation complex is ready to start transcription. How does the enzyme get started A component of TFIID, again a multi-subunit complex TFIIH, unwinds the DNA and phosphorylates serine-5 of the C-terminal tail (CTD) of the largest polymerase subunit (Rpbl). Serine-5 phosphorylation and phosphorylation of serine-2 (by pTEFb) are required to release the enzyme from the other components of the initiation complex and to start RNA synthesis. [Pg.1225]

Nickel(IV) complexes react with dimethyl sulphoxide in acidic solution to give the sulphone and nickel(II) ions. The kinetics of this reaction have been studied and found to be very complex in nature. The reaction probably proceeds by initial complexation of the dimethyl sulphoxide to the nickel(IV) species followed by electron transfer and oxygen atom transfer producing the observed products149. [Pg.985]

The exact mechanism has still not been completely worked out. Opinions have been expressed that it is completely intermolecular, completely intramolecular, and partially inter- and intramolecular. " One way to decide between inter- and intramolecular processes is to run the reaction of the phenolic ester in the presence of another aromatic compound, say, toluene. If some of the toluene is acylated, the reaction must be, at least in part, interraolecular. If the toluene is not acylated, the presumption is that the reaction is intramolecular, though this is not certain, for it may be that the toluene is not attacked because it is less active than the other. A number of such experiments (called crossover experiments) have been carried out sometimes crossover products have been found and sometimes not. As in 11-14, an initial complex (40) is formed between the substrate and the catalyst, so that a catalyst/substrate molar ratio of at least 1 1 is required. [Pg.726]

There has been much controversy about whether the initial complex in the... [Pg.1202]

Crystal structures of the NS5B polymerase alone and in complexes with nucleotide substrates have been solved and applied to discovery programs (Ago et al. 1999 Bressanelli et al. 2002 Bressanelli et al. 1999 Lesburg et al. 1999 O Farrell et al. 2003). From these studies, HCV polymerase reveals a three-dimensional structure that resembles aright hand with characteristic fingers, palm, and thumb domain, similar to the architectures of the RNA polymerases of other viruses. However, none of these experimental structures contained the ternary initiation complex with nu-cleotide/primer/template, as obtained with HIV RT. Accordingly, HCV initiation models have been built using data from other viral systems in efforts to explain SAR (Kozlov et al. 2006 Yan et al. 2007). [Pg.32]

The first examples of cationic exchange of bis(oxazoline)-metal complexes used clays as supports [49,50]. Cu(II) complexes of ligands ent-6a, 6b, and 6c (Fig. 15) were supported on three different clays laponite (a synthetic clay), bentonite, and montmorillonite KIO. The influence of the copper salt from which the initial complexes were prepared, as well as that of the solvent used in the cationic exchange, was analyzed. [Pg.174]

In contrast to most mRNAs, which become untranslatable after a temperature downshock, cold shock mRNAs possess a mechanism to form the translation initiation complex at low temperature without cold shock ribosomes. A close inspection of the mRNAs of class I cold shock proteins reveal that they are equipped with an extra ribosome-binding site called the downstream box located within the coding region of their transcript [130]. It would be interesting to know whether introduction of this downstream box into a cellular mRNA would convert it into a transcript which can be transcribed immediately after a cold shock. In the case of the cspA mRNA it has been shown that in the absence of the downstream box the initiation complex cannot be formed at low temperature during the accHmation phase [131]. [Pg.27]

The processes of DNA and RNA synthesis are similar in that they involve (1) the general steps of initiation, elongation, and termination with y to 3 polarity (2) large, multicomponent initiation complexes and (3) adherence to Watson-Crick base-pairing rules. These processes differ in several important ways, including the... [Pg.341]

TBP binds to the TATA box in the minor groove of DNA (most transcription factors bind in the major groove) and causes an approximately 100-degree bend or kink of the DNA helix. This bending is thought to facilitate the interaction of TBP-associated factors with other components of the transcription initiation complex and possibly with factors bound to upstream elements. Although defined as a component of class II gene promoters, TBP, by virtue of its association with... [Pg.350]

Initiation of protein synthesis requires that an mRNA molecule be selected for translation by a ribosome. Once the mRNA binds to the ribosome, the latter finds the correct reading frame on the mRNA, and translation begins. This process involves tRNA, rRNA, mRNA, and at least ten eukaryotic initiation factors (elFs), some of which have multiple (three to eight) subunits. Also involved are GTP, ATP, and amino acids. Initiation can be divided into four steps (1) dissociation of the ribosome into its 40S and 60S subunits (2) binding of a ternary complex consisting of met-tRNAf GTP, and eIF-2 to the 40S ribosome to form a preinitiation complex (3) binding of mRNA to the 40S preinitiation complex to form a 43S initiation complex and (4) combination of the 43S initiation complex with the 60S ribosomal subunit to form the SOS initiation complex. [Pg.365]

The binding of the 60S ribosomal subunit to the 48S initiation complex involves hydtolysis of the GTP bound to elF-2 by elF-5. This teaction tesults in telease of the initiation factots bound to the 48S initiation complex (these factots then ate tecycled) and the tapid association of the 40S and 60S subunits to fotm the 80S ribosome. At this point, the met-tRNA is on the P site of the ribosome, ready for the elongation cycle to commence. [Pg.367]

The simplest interpretation of equation (15) would assume a nucleophilic attack on Co(III) by OH-. This, however, would put OH in an extraordinary category of nucleophilicity. Garrick was the first to note that an alternative explanation for the role of OH was available. In the alternative, the conjugate base of the initial complex ammine is presumed to be formed in small amount and to function as the actual reactive species... [Pg.16]
