The initial product resulting from the action of a lipoxygenase is a hydroperoxy acid. This is quickly followed by reduction to a hydroxyacid by peroxidase. Some common anti-inflammatory drugs have been shown to cause accumulation of 12-HPETE in platelets by inhibition of the enzymatic reduction into 12-HETE [186,188]. Among the drugs showing this effect are aspirin, indomethacin, sodium salicylate and phenylbutazone. [Pg.157]

Indomethacin, sodium salicylate and Budesonide Discs In-vitro studies revealed there was complete sodium salicylte release within 120 min. While Budesonide and Indomethacin showed minimal release for 10 h. [144]... [Pg.336]

Replacement of chlorine on the pendant benzoyl group by azide is apparently consistent with antiinflammatory activity. Acylation of indomethacin intermediate with p-nitrobenzoyl chloride leads to the corresponding amide (7). Saponification ( ) followed by reduction of the nitro group gives the amine 9. The diazonium salt (10) obtained on treatment with nitrous acid is then reacted with sodium azide there is thus obtained zidomethacin (11). [Pg.166]

The FT-Raman spectra of a range of drugs (theophylline, indomethacin, diclofenac, and promethazine) in several polymers (sodium alginate, hydroxy-propylmethylcellulose, and polyethylene glycol) have been obtained [56,57]. In these studies, the linearity of response of Raman scattering to species concentration was exploited to analyze diclofenac at concentrations of 0.01-6.0% w/w... [Pg.82]

Eideriy Age appears to increase the possibility of adverse reactions to NSAIDs. The risk of serious ulcer disease is increased this risk appears to increase with dose. Ketorolac is cleared more slowly by the elderly use caution and reduce dosage. Pregnancy Category B (ketoprofen, naproxen, naproxen sodium, flurbiprofen, diclofenac, fenoprofen, ibuprofen, indomethacin, meclofenamate, sulindac). [Pg.939]

Lithium intoxication can be precipitated by the use of diuretics, particularly thiazides and metola-zone, and ACE inhibitors. NSAIDs can also precipitate lithium toxicity, mainly due to NSAID inhibition of prostaglandin-dependent renal excretion mechanisms. NSAIDs also impair renal function and cause sodium and water retention, effects which can predispose to interactions. Many case reports describe the antagonistic effects of NSAIDs on diuretics and antihypertensive drugs. The combination of triamterene and indomethacin appears particularly hazardous as it may result in acute renal failure. NSAIDs may also interfere with the beneficial effects of diuretics and ACE inhibitors in heart failure. It is not unusual to see patients whose heart failure has deteriorated in spite of increased doses of frusemide who are also concurrently taking an NSAID. [Pg.258]

Indomethacin is defined as the Form I crystalline, non-solvated free acid moiety of the compound unless otherwise noted. Indomethacin as described below can exist as several crystalline forms but has been used in pharmaceutical preparations principally as Form I and less frequently as the crystalline sodium trihydrate. [Pg.213]

Indomethacin has also been synthesized from sodium p-methoxy-phenylhydrazine sulfonate and from saffrole(33,34). [Pg.227]

Assessment of indomethacin powder had been specified in the U.J5. XIX and the British Pharmacopeia, 1980 as a back titration with hydrochloric acid after alkaline hydrolysis(13,14). This method can attain a precision of + 0.8%(62). A direct titration for tablets and capsules is described using sodium hydroxide. If performed rapidly with phenolphthalein indicator, a precision of +0.3% is attainable. 8 The latter procedure serves to differentiate ester formation as well as hydrolysis products from intact material. The presence of parachlorobenzoic acid and 5-methoxy-2-methylindole-3-acetic acid are cause for positive error. [Pg.234]
