Immunosuppressant signal transduction

FKBP12 is a member of immunophilin family that has prolyl isomerase activity and is related to the cyclophi-lins in function. FKBP12 binds immunosuppressant molecule FK506 (tacrolimus). The FBKP-FK506 complex inhibits calcineurin, a protein phosphatase, thus blocking signal transduction in the T-lymphocyte... [Pg.507]

The monoclonal antibodies used as immunosuppressive agents in tissue transplantation include muromonoab-CD3, daclizumab and basiliximab. Muromonoab-CD3 binds to a specific site on CD3 receptors and interferes with the ability of the TCR to bind the antigen and also inhibits CD3 receptor-dependent signal transduction mechanisms, all of which result in immune suppression. Both daclizumab and basiliximab are monoclonal antibodies directed against IL-2 receptors and consequently inhibit IL-2-dependent responses after tissue transplantation, resulting in immune suppression. The monoclonal antibodies used as immunosuppressive agents are described in detail in Chapter 5. [Pg.102]

Natural products, in particular those produced by microorganisms, have proved to be a rich source of useful signal transduction inhibitors with a variety of mechanisms of action. They continue to be of great interest as a source of new signal transduction inhibitors with potential for therapeutic use in the areas of immunosuppression, oncology and infectious disease. [Pg.87]

Signal transduction by IL-10 is stfll unknown but likely to involve the JAK family of kinases. The signal transduction cascades initiated by IL-10 ligation to its cognate receptor seem to rapidly and transiently stimulate phosphatidylinos-itol 3-kinase activity associated with the p85 subunit of the enzyme. IL-10 also activates p70 S6 kinase that is inhibited by the immunosuppressive drug rapamycin. ... [Pg.679]

The exact signal transduction mechanism that triggers the immunosuppressive response in T-cells, however, remained un-... [Pg.100]
