Another means of moving beyond pure protein preparations to high-throughput characterization of proteomes is to enrich for phosphopeptides from complex mixtures by metal affinity chromatography (Andersson and Porath, 1986). Using this method, protein mixtures are proteolyzed to create peptides and phosphorylated peptides are enriched by metal affinity chromatography and subsequently identified by mass spectrometry. This method is limited, however, because in many cases phosphopeptides absorb poorly or nonphosphorylated peptides absorb nonspecifically to the metal affinity resins (Ahn and Resing, 2001). [Pg.19]

In addition to quantitative crystallinity data, IR and Raman have been proven valuable tools to extract information on chain conformation in the three major phases [112-114], local order in amorphous polymers [115,116] high throughput characterization [117] and structural and polymorphic changes on heating and cooling semi-crystalline polymers [118-120]. [Pg.266]

The next section describes the utilization of //PLC for different applications of interest in the pharmaceutical industry. The part discusses the instrumentation employed for these applications, followed by the results of detailed characterization studies. The next part focuses on particular applications, highlighting results from the high-throughput characterization of ADMET and physicochemical properties (e.g., solubility, purity, log P, drug release, etc.), separation-based assays (assay development and optimization, real-time enzyme kinetics, evaluation of substrate specificity, etc.), and sample preparation (e.g., high-throughput clean-up of complex samples prior to MS (FIA) analysis). [Pg.158]

The present workflow includes two complementary high-throughput characterization methods X-ray diffraction (XRD) for structural information and scanning electron microscope/energy dispersive X-ray (SEM/EDX) analysis for quantitative compositional information. Automated rapid-serial XRD and EDX analyses were performed using programmable xy-stages in combination with suitable software. [Pg.277]

Ujiie et al. [204] fabricated quartz chips for NCE and reported the separation of rhodamine B and sulforhodamine at 14.4 and 66.6 cm separator lengths. The buffer was 20 mM phosphate buffer at 2kV applied voltage and the separation was achieved in 70 seconds. Wakida et al. [205] reported a high throughput characterization for dissolved organic carbon in environmental waters within 2 minutes using NCE. The authors collected water samples from 10 sampling points at the Hino River that flows into Lake Biwa. Shin et al. [206] described NCE (PDMS) with fluorescence detection for analyses of atrazine. [Pg.231]

Open-access MS systems provide an effective means for maintaining the high-throughput characterization of synthetic compounds. These systems offer an efficient laboratory- to bench-scale integration of sample generation and analysis activities. Advances in analytical instrumentation and electronic communication have also played a major role in the emergence and acceptance of MS as a front-line tool for structure characterization. [Pg.3425]

A combined ESI-APCI source is available from several instmment manufacturers. It was developed for high-throughput characterization of combinatorial hbraries [103], The system uses the existing API source, and allows alternating ESI and APCl within one chromatographic ran. [Pg.127]

Initially, high-throughput characterization of combinatorial libraries was developed for and performed with single quadmpole instmments, providing unit-mass resolution and nominal molecular-mass determination. The application of high-resolution mass spectrometers, such as time-of-flight (TOF) and FT-ICR-MS, further enhances the power of the approach [22]. [Pg.239]
