The third observation that supported a central role for DNA in cancer came from the study of chromosomes in human leukemia cells. Karyotyping is a cytological technique used to characterize the gross structure of chromosomes in mitotic cells. Researchers noticed that in cells from one type of leukemia, chronic myelogenous leukemia, there was almost always a chromosome rearrangement between chromosomes 9 and 22. This reciprocal translocation led to identification of a small 22 9 chromosome which came to be called the Philadelphia chromosome because it was discovered at the Wistar Institute in Philadelphia. Importantly, the karyotype of nonleukemic cells from patients with this form of leukemia is normal. [Pg.880]

In vitro studies have shown that GNE/MNK is able to interact with factors such as the coUapsin response mediator protein 1 and the promyelocytic leukemia zinc-finger protein [35], but none of these molecular partners has been proven so far to be involved in the pathogenic cascade of h-IBM muscle. More recently, GNB/MNK has been demonstrated to physically interact also with a-actinin 1, and these two proteins partially colocalize in the sarcomere of mature muscle fibers. However, in vitro, no gross difference has been observed between the interaction of a-actinin 1 with wild-type GNB/MNK and mutated GNB/MNK [36]. Although this line of evidence provides novel interesting clues on additional roles of GNB/MNK outside of the sialic acid biosynthetic pathway, it remains to be determined how even a minor impairment of the interaction between a-ac-tinin 1 and GNB/MNK, as hypothesized in h-IBM, may impact the viability of muscle fibers. [Pg.202]
