In vitro and animal studies of the eifects of phytoestrogens on established tumors have raised some concerns that may be relevant to women at high risk of breast cancer. Although some studies have shown inhibitory eifects of phytoestrogens (e.g., Hawrylewicz et al., 1995), others have reported increased carcinogenesis in rodents (e.g., Ju et al., 2001). Of particular concern is a recent report that dietary genistein negates the inhibitory effect of tamoxifen in the athymic nude mouse model (Ju et al., 2002).

All isoflavones [daidzein (27), genistein (28)] and isoflavanones (17, 86-93) did not stimulate nitric oxide production by mouse macrophage-like Raw 264.7 cells, but almost completely inhibited NO production by the lipopolysaccharide (LPS)-activated Raw 264.7 cells. ESR spectroscopy showed that secundifloran (88) and secundiflorol D (90), which showed the most inhibitory effect for NO production, efficiently scavenged the superoxide anion and NO radicals. From these results, it is suggested that the inhibition of macrophage NO production by these isoflavanones might, at least in part, be explained by their radical scavenging or reduction activity (Fig. 17) (Tables 3, 4) [16]. 

Preincubation of the cells with genistein invariably blocked the Ca + response evoked by either phenylephrine or serotonin (Figs. 4C and 4D). Though not shown, the inhibitory effects of genistein were reversible as replacement of the medium with genistein-free PSS resulted in recovery of responsiveness to both phenylephrine and serotonin. Tyrosine kinases have been implicated in Ca +-sensitization largely as the result of the inhibitory effects of tyrosine kinase inhibitors, genistein and vanadate, and the correlation between tyrosine phosphorylation and vanadate- or agonist-induced contractions (Di Salvo et al. 1993a,b, 1994, 1997 Steusloff et al. 1995). However, the tyrosine-phosphorylated proteins have yet to be directly linked to a known contractile regulatory mechanism i.e., MLCK, SMPP-IM) and evaluation of the relationships between tyrosine phosphorylation and modulation of Ca +-sensitization of contraction in intact muscle is somewhat complicated by concurrent changes in cytosolic Ca (Di Salvo et al. 1994,1997), possibly as the result of activation of PLC-y (Marrero et al. 1994) or Gaq.n (Umemori et al. 1997).

Genistein is one of the best known and studied of the isoflavones. It activates the three isoforms, a, 5, and y, of PPARS, binding more readily with ERp than ERa. It also acts as an antioxidant. It has inhibitory effects on prostate, cervical.
