Bromelain is the collective name for closely related proteolytic enzymes found in tissues of the plant family Bromeliaceae, of which pineapple, Ananas comosus, is the best known. Two distinct types of pineapple bromelain are recognized stem bromelain (EC 3.4.22.32), the major proteolytic cysteinyl protease in pineapple stem, and fruit bromelain (EC 3.4.22.33), formerly called bromelin, which is the major proteolytic component in pineapple fruit [1]. Stem bromelain has been known under a variety of synonyms, namely, pineapple stem bromelain, EC 3.4.4.24 (formerly), EC 3.4.22.5 (formerly), included in EC 3.4.22.4 (formerly), ananase, bromelase, pinase, extranase, traumanase, pineapple enzyme, and juice bromelain. [Pg.131]

A detailed study concerning the occurrence of different proteolytic enzymes in crude stem and fruit bromelain was carried out by Ota et al. [15]. Fractionation of crude stem bromelain and characterization of the components revealed six proteolytically active components (SBB 1-5 and SBA) [15,16]. All active fractions were analyzed by SDS PAGE and migrated as a single band with molecular weights of 27,000 for SBB 1-3 and about 23,000 for SBB 4 and 5. A potent cysteine proteinase inhibitor PCPI 8.3 was isolated from potato tubers. The inhibitor has a broad inhibitor spectrum, including the bromelain enzymes, which are not inhibited by cystatin. PCPI shows a Ki value of 190 nM for stem bromelain, 33 nM for fruit bromelain, 0.06 nM for ananain, and 3.3 nM for papain [78]. It is proposed that the differences of inhibitory spectrum between PCPI and the cystatins may be those of distinct superfamilies of cysteine proteinase inhibitors. [Pg.141]
