If this procedure is followed, then a reaction order will be obtained which is not masked by the effects of the error distribution of the dependent variables If the transformation achieves the four qualities (a-d) listed at the first of this section, an unweighted linear least-squares analysis may be used rigorously. The reaction order, a = X + 1, and the transformed forward rate constant, B, possess all of the desirable properties of maximum likelihood estimates. Finally, the equivalent of the likelihood function can be represented b the plot of the transformed sum of squares versus the reaction order. In the fourth century B.C., the influential Greek philosopher Aristotle (384—322 B.c.) described the composition and behavior of matter in terms of the four qualities shown in Figure 3.1 hot, cold, moist, and dry. Although we know today that Aristodes model is wrong, it was nonetheless a remarkable achievement for its day, and people using it in Aristotle s time found it made sense. When pottery was made, for example, wet clay was converted to ceramic because the heat of the fire drove out the moist quality of the wet clay and replaced it with the dry quality of the ceramic. Likewise, warm air caused ice to melt by replacing the dry quality characteristic of ice with the moist quality characteristic of water. [Pg.76]

Before the laser, the light sources used in infrared, visible, and ultraviolet spectroscopy were heated solids or gas discharge tubes. These sources are based on spontaneous emission. A selective, sensitive, and rapid hydrophilic interaction liquid chromatography with electrospray ionization tandem mass spectrometry was developed for the determination of donepezil in human plasma [32], Donepezil was twice extracted from human plasma using methyl-ferf-butyl ether at basic pH. The analytes were separated on an Atlantis HILIC Silica column with the mobile phase of acetonitrile ammonium formate (50 mM, pH 4.0) (85 15, v/v) and detected by tandem mass spectrometry in the selective reaction monitoring mode. The calibration curve was linear (r = 0.9994) over the concentration range of 0.10-50.0 ng/ ml and the lower limit of quantification was 0.1 ng/ml using 200 /d plasma sample. The CV and relative error for intra- and inter-assay at four quality control levels were 2.7% to 10.5% and —10.0% to 0.0%, respectively. There was no matrix effect for donepezil and cisapride. The present method was successfully applied to the pharmacokinetic study of donepezil after oral dose of donepezil hydrochloride (10 mg tablet) to male healthy volunteers. [Pg.141]

In addition to the four elements of earth, water, air, and fire, Aristotle also included the four qualities and the four causes. The four qualities were set in contrasting pairs hot/cool, wet/dry. At the simplest level, the qualities linked the four elements the combination of water and air was wet, while fire and earth was dry. There was a deeper purpose to the qualities, however. They were necessary to explain the observed condition of physical objects, since the simple proportions of each element was not sufficient. For example, blood could not simply be described as a particular mixture of water and fire elements, since it was qualitatively different from bile but must have some of the same elemental makeup. [Pg.15]
