Given the above preclinical observations, patients in a clinical study were monitored closely during treatment (including measurement of testosterone, luteinizing hormone, follicle-stimulating hormone levels, alkaline phosphatase, and serum vitamin A concentrations). This phase I clinical study of TAG-101 was conducted to determine the safety, toxicity, and pharmacokinetics of this agent in patients with advanced cancer. Currently, the drug is in a phase I/II clinical trial for advanced hepatocellular carcinoma.

In prepubertal boys, but not in older boys or adult men, hypothyroidism is associated with a high incidence of increased testicular growth (macroorchidism) that is not accompanied by any change in testosterone levels. Elevated follicle-stimulating hormone levels are found in most cases, but there is no testicular maturation. The mechanism of this disorder is not known. 

Seven men who were exposed for 6 hours/day for 5 days to 616 mg/m of vaporized white spirits (with a composition of 99% alkanes, as compared to 30-50% alkanes in Stoddard solvent) had a decrease (p<0.05) in serum follicle-stimulating hormone levels at 24 and 96 hours after the initiation of exposure as compared to pre-exposure levels (Pedersen and Cohr 1984b). This change did not correspond to blood or adipose levels of white spirits. No tests of reproductive function were performed. In another study, 11 men in a printing factory were occupationally exposed to a wide variety of solvents, including 294 mg/m of white spirits for 1-17 years. Sperm counts, motility, and morphology were monitored for 2 months, and all values were normal (Tuohimaa and Wichmann 1981).

Disappearance of the negative feedback control of hormones leads to uncontrolled hormone secretion. For example, ovarian failure—as it happens in menopause—is caused by the luteinizing hormone and follicle-stimulating hormone levels increase due to the lack of negative feedback effects of the gonadal hormones. 

In women aged 35 to 70 with at least one first-degree relative with breast cancer, administration of 40 mg of red clover isoflavones daily for 3 years did not result in any significant differences in breast density, endometrial thickness, serum cholesterol, follicle-stimulating hormone levels, or bone mineral density, as compared to placebo. In postmenopausal women, some differences in bone marker levels were seen between active and placebo groups at 6 and 12 months (Powles et al. 2008).
