In some instances, flow cytometry assays are a superior alternative to conventional procedures for the determination of equilibrium binding constants (Stein et al., 2001). In contrast to assays that employ radiolabelled ligands, which measure population mean values for binding constants, flow cytometry methods can measure those values in individual cells, revealing heterogeneity in receptor expression within a population of cells or membrane vesicles. Furthermore, small samples can be characterized in a short period of time (hours). This approach to receptor-binding analysis may be limited only by the availability of a properly characterized fluorescent ligand. [Pg.312]

Owing to its applications in homeland security, sterilization validation, and astrobiology, bacterial spore detection has become a hot field. However, direct detection of bacterial spores can be challenging for the same reasons that make endospores difficult to irradicate. The tough spore coat is impermeable to staining techniques, so most microscopy and flow cytometry methods are not useful. Endospores are also highly resistant to lysis, meaning that common DNA extraction protocols are difficult to perform. The lack of measurable metabolism renders microcalorimetry and cellular respiration techniques ineffective. [Pg.12]

Treatment results in childhood acute leukemia continue to be improved due to new therapeutic and diagnostic methods. An optical microscopy method with specific staining allows to classify acute leukemia into lymphoblastic and myeloid types and discriminate its subtypes according to principals of FAB-classification. A flow cytometry method with monoclonal antibodies staining allows us to discriminate different immunophenotypic subtypes of acute... [Pg.524]
