You can retrieve an intermediate structure from the output log file manually. Alternatively, you may use the Geom=(Check,Step=n) keyword to retrieve the structure corresponding to step n from a checkpoint file. 

In addition to dtrate, there are other addic intermediates of file TCA cyde and several of these are used in industry. Even though some have been produced by fermentation, production is currently from other, cheaper sources.

Intermediates don t often reach the marketplace but do require a PMN under TSCA. Similarly, many chemicals apparently had PMNs filed to be ready for manufacture/marketing but then failed to make it through the business decision process for other reasons.

The Chemical Manufacturers Association (CMA) had reached similar conclusions about a year ago and filed a petition with EPA suggesting that there was a strong case to be made for exemptive relief under Section 5 (h)(4) of TSCA for many polymers, site-limited intermediates, and chemicals produced in volumes of less than 25,000 pounds per year. (An examination of the effect of PMN costs at various prices and levels of production reveals that the PMN cost, as a percentage of total cost per pound of product, generally rises most rapidly as output falls below 25,000 pounds. See the Appendix for a more detailed discussion.) It is CMA s firm belief that exemption could be granted in terms that would operate to assure "no unreasonable risk" to the public in terms of either health or the environment. 

Reproducibility is the third and final portion of precision testing. Here, samples are prepared and compared between testing sites. This usually occurs at the time of technology transfer. Samples are prepared in a similar manner between the two sites and are compared to a previously agreed-upon set of acceptance criteria. As per the ICH, reproducibility studies are not a part of submission filings, but should be performed as a confirmation of the ability of each testing site to perform the method reliably. An executed reproducibility study can be used in place of intermediate precision for the validation of a method, although there is no problem if both validation parameters are evaluated. 

The most convenient method is ultrasonic treatment with a subsequent solvent rinse of the surface. Other intermediate procedures are abrasive scrubbing, filing, or detergent cleaning. 

Recently, Hu and co-workers, at Pfizer, filed a patent application on a process to prepare pregabalin (2) via enzymatic resolution (Hu et al., 2005). The Pfizer researchers investigated a large number of commercially available hydrolases to catalyze the hydrolysis of (3-cyanodiester 59, an intermediate from the malonate route (Scheme 16.16), to form (35)-3-cyano-2-ethoxcarbonyl-5-methylhexanoic acid potassium salt (71)... 

The authors of this work are obviously mindful of the potential commercial exploitation of these techniques as is evidenced by the filing of patent protection by Morita et al. [24], in which they claim the route towards organic compounds containing fluorine useful as intermediate raw material of drugs, pesticides, and liquid crystals.
