In order to better understand the pathophysiology associated with these various forms of pulmonary edema, a review of the morphology associated with the capillary-alveolar—intestinal interlace is useful (Fig. 6). From this review it can be seen that fluid in pulmonary capillaries is separated from the alveolar interstitial tissue by the capillary endothelial cells and the capillary basement membrane (commonly called the endothelial barrier). The alveolar surface is separated from the interstitial space by the alveolar-airway barrier, which consists of the alveolar basement membrane, alveolar epithelium, and a layer of pulmonary surfactant within the alveolus. As described previously, the alveolar interstitial tissue is made up of connective tissue (elastin and collagen), fibronectin, and mucopolysaccharides. The interstitial space also contains the pulmonary lympatic system, which functions to drain proteins, large particulate matter, and excess fluid away from the tissue space and to return them to the blood. [Pg.360]

The Boyden chamber is a simple apparatus used to test for chemotaxis, especially of leukocytes. It can also be used to assess tumor cell transmigration across an endo-thehal monolayer in vitro. It consists of two compartments separated by a MiUi-pore filter (3-8 pm pore size). A chemotac-tic factor is placed in one compartment, and a gradient develops across the thickness of the filter (ca. 150 pm). Cell movement into the filter is measured after an incubation period less than the time taken for the gradient to decay. Cell motility can be measured in Boyden chambers containing filters precoated with different materials, for example fibronectin or fibronectin fragments. The method, when apphed to malignant and non-mahgnant cell hnes, shows that the variable invasive potentials of these cells correlate with their abihty to disrupt the endothelial cell monolayer. [Pg.643]

The control sample of horse cells in isoton showed somewhat weaker adhesion than the sample shown in Table 12.2. Such variation was found to be common in different samples of horse blood. Differences between animals in type, age, etc., and also in blood cell conditioning had a distinct influence which will be described in separate papers. It is evident from the results that glutaraldehyde reduced the adhesion by about 25% whereas fibronectin increased the adhesion by 10% and papain by 20%, changes which were comparable with the effects seen on human red cells but disappointingly small compared with the effects anticipated. [Pg.296]
