Glyoxylic acid ethyl ester ethyl alcoholate Manufacturing Process 

7 g of benzotetronic acid are dissolved in 750 cc of water at boiling temperature and thereafter 10.5 g of glyoxylic acid ethyl ester ethyl alcoholate are added. After a short while the liquid becomes turbid and gradually a white deposit is separated. The deposit is filtrated and dried in vacuo. The melting point is 172°C to 174°C after recrystallization from methyl alcohol 153°Cto 154°C. 

The crude product is dissolved in sodium lye, filtrated by means of animal charcoal precipitated by means of hydrochloric acid,and recrystallized from methyl alcohol. The melting point is 153°C to 154°C.

Ethyl biscoumacetate Ethyl biscoumacetate, the ethyl ester of fcw-(4-hydroxy-3-conmarinyl)-acetic acid (24.1.9), is synthesized analogously from 4-hydroxycoumarine, but using ethylglyoxylate, its semiacetal or glyoxylic acid instead of formaldehyde [10-13]. 

In 10 of 14 patients taking dicoumarol or phenindione who also took corticotropin for 4—9 days, there was a small increase in the effects of the anticoagulants (31). One patient taking ethyl biscoumacetate developed bleeding after taking corticotropin 20 mg/day for 3 days (32). However, in contrast, reduced efficacy of ethyl biscoumacetate has been described (33).

Since dimeticone is a surfactant, one might expect it to enhance the absorption of drugs, and there are some reports that this happens with ethyl biscoumacetate (17).

The coumarins were first discovered in Wisconsin, when bleeding in cattle was found to be due to the consumption of bruised sweet clover in the 1920s (1). The causative agent, dicoumarol, was isolated in 1940, and a range of related compounds was then synthesized, the most popnlar of which proved to be warfarin (named after the Wisconsin Alumni Research Foundation). Other conmarins that have been used are acenocoumarol (nicoumalone), bishydroxycoumarin, dicoumarol, ethyl biscoumacetate, and phenproconmon. 

The coumarin anticoagulants include dicumarol, warfarin sodium (coumadin sodium), warfarin potassium (Athrombin-K), acenocoumarol, and phenprocouman. Phenprocouman, acenocoumarol, and ethyl biscoumacetate are not generally available in the United States but are prescribed in Europe and elsewhere. Phenprocouman (Marcumar) has a longer plasma half-life (5 days) than warfarin, as well as a somewhat slower onset of action and a longer duration of action (7 to 14 days). It is administered in daily maintenance doses of 0.75 to 6.0 mg. By contrast, acenocoumarol (Nicoumar-lone Sinthrome) has a shorter half-life (10 to 24 hours), a more rapid effect on the prothrombin time, and a shorter duration of action (2 days). The maintenance dose is 1 to 8 mg daily. Ethyl biscoumacetate (Tromexane) has a very short half-life of 2 to 3 hours and is seldom used.
