Chlorogenic and caffeic acids inhibit the activation of hyaluronidase, which is known as one of the enzymes involved in allergic effects [79], migration of cancer cells [80], inflammation [81, 82], and increased permeability of the vascular system. The superoxide anion radical scavenging activity of chlorogenic acids correlates linearly to the hyaluronidase-inhibitory activity. (3-Hexosaminidase release from rat basophilic leukemia cells induced by antigen has been inhibited by chlorogenic and caffeic acids. An important example of PLP-dependent amino acid decarboxylation is the conversion of histidine into histamine. Histamine is often involved in human allergic responses, e.g. to insect bites or pollens. Stress stimulates the action of the enzyme histidine decarboxylase and histamine is released from mast cells. Topical antihistamine creams are valuable for pain relief, and oral antihistamines are widely prescribed for nasal allergies such as hay fever. Major effects of histamine include dilation of blood vessels, inflammation and swelling of tissues, and narrowing of airways. In serious cases, life-threatening anaphylactic shock may occur, caused by a dramatic fall in blood pressure. [Pg.601]

Pemetrexed is chemically similar to folic acid. It inhibits three enzymes used in purine and pyrimidine synthesis - thymidylate synthetase, dihydrofolate reductase, and glycinamide ribonucleotide formyl transferase. By inhibiting the formation of precursor purine and pyrimidine nucleotides, pemetrexed prevents the formation of DNA and RNA. In 2004 it was approved for treatment of malignant pleural mesothelioma and as a second-line agent for the treatment of non-small cell lung cancer. Adverse effects include gastrointestinal complaints, bone marrow suppression, alopecia, allergic and neurotoxic reactions. [Pg.452]
