Elementary Precautions

One of the problems one may faee when analysing quadruplexes, is that, onee formed, some structures are so diffieult to disrupt that even ineubating for 

10-30 min at 100°C does not lead to signifieant quadruplex unfolding, even in the presence of 7 M urea. When undesired quadruplex formation is observed before a kinetic experiment, the addition of limited amounts of sodium hydroxide followed by HCl neutralization is a convenient and fast method to disrupt these complexes but results in a net increase in salt eoncentration. As a consequence, we suggest using a 0.1 M LiOH solution to unfold these oligomers. 

Several controls may be performed to confirm that the folded structure indeed corresponds to a quadruplex. The cation dependency of its stability (which follows a K Na Li preference order) is a good indieation. One may also demonstrate that the thermal difference spectra or circular dichroism spectra of all these structures are in agreement with the formation of quadruplexes. Furthermore, non-denaturing gel electrophoresis may confirm that a single, major retarded (for hi- and tetramolecular) band is observed when the oligonue-leotides are pre-incubated at high concentration in the presence of NaCl or KCl.

CAUTION. Fluorine is a strong oxidizer and a very corrosive material. An appropriate vacuum line made from copper or Monel in a well-ventilated area should be constructed for working with this element. The reactions themselves were carried out in Teflon vessels. Different clarified juices must often be assembled when filling a high-capacity fermenting tank. This operation requires several elementary precautions. Precautions for Elementary and Junior/Senior High Schools...

When Raman bands are overlapped some additional precaution must be taken.
