Drugs fluorescamine

Many derivatization techniques, based on chemical reactions or even heat and chemical reactions, can be applied to TLC or HPTLC to specifically determine some compounds or groups of compounds this is specific to TLC techniques. For instance, exposure to fluorescamine in acetone wiU convert sulfonamide antibiotics into fluorescent derivatives which can be visualized under UV radiation [8]. Other techniques are reported and, as suggested earher, the reader should refer to the latest edition of the Official Methods of Analysis pubhshed by the Association of Official Analytical Chemists (Arlington VA, U.S.A.) for complete details about aU the available techniques for the routine analysis of drugs. [Pg.1163]

Sulfonamides are used for the prevention and treatment of human infectious diseases and also as growth promoters in swine. It is necessary to monitor pork products for drug residues of sulfonamides. Various sulfonamides were separated on Whatman LKCigF layers in a methanol-0.5 M NaCl (50 50) mobile phase spots were detected with UV light (366 nm) after spraying with fluorescamine reagent followed by 0.5% triethanolamine in chloroform to stabilize fluorescence. The Rp values of the sulfonamides were as follows sulfabromomethazine, 0.19 sulfadimethoxine, 0.39 sulfamethazine, 0.59 sulfathiazole, 0.76 sulfaguanidine, 0.90 (Whatman TLC Technical Series, Volume 3). [Pg.438]

Thomas et al. (1981) quantified sulfamethazine in swine tissues at 0.1 ppm by densitometry of the fluorescamine derivative on preadsorbent silica gel layers. Tissue was extracted with ethyl acetate, and the drug was back extracted into methylene chloride, and concentrated. Samples and standards were developed with chloroform-tertiary butanol (80 20). [Pg.438]

Other solvent systems and detection reagents are recommended for the identification and confirmation of drugs and their metabolites. These include ethyl acetate-methanol-concentrated ammonium hydroxide (90 7 3) with detection by fluorescamine, diphenylcarbazone, and iodoplatinate reagents for D-amphet-amine, barbiturates, and acidic drugs (e.g., phenobarbital, Dilantin, Mellaril, Valium, chlorpromazine, Lomotil, and Talwin) and ethyl acetate-methanol-concentrated ammonium hydroxide-water (85 13.5 0.5 1.0) with detection by... [Pg.443]
