4- Diphenylhydantoin

Many hydantoins are endowed with significant pharmacological activities as highlighted by 5,5-diphenylhydantoin (Dilantin ), an anticonvulsant and antiepileptic discovered by Parke-Davis in 1940 s. Despite the lapse of more than half a century, Dilantin still plays an important rote in modem medicine. Meanwhile, another anticonvulsant 15 was synthesized from 9,10-dimethoxy-l,3,4,6,7,llb-hexahydro-pyrido[2,l-a]isoquinolin-2-one (14) under the standard Bucherer-Lieb variation in a 2 1 water-ethanol solution (15a 15b = 8 l). ... [Pg.268]

Hydantoin analogs in this series 72) were prepared. Most of these compounds, however, were found less active as anticonvulsants than the corresponding phenyl derivatives. Only a few were found to have the same order of activity as 5,5-diphenylhydantoin. [Pg.128]

Urea may be recognised as a structural unit in 5,5-diphenylhydantoin (57) and it constitutes one of the reagents in the synthesis. Although it is not obvious, the appropriate reagent equivalent corresponding to the accompanying synthon is in fact benzil. [Pg.1152]

Place 5.3 g (0.025 mol) of benzil (Expt 6.143), 3.0 g (0.05 mol) of urea, 15 ml of 30 per cent aqueous sodium hydroxide solution and 75 ml of ethanol in a 100-ml round-bottomed flask. Attach a reflux condenser and boil under reflux using an electric heating mantle for at least 2 hours. Cool to room temperature, pour the reaction product into 125 ml of water and mix thoroughly. Allow to stand for 15 minutes and then filter under suction to remove an insoluble by-product. Render the filtrate strongly acidic with concentrated hydrochloric acid, cool in ice-water and immediately filter off- the precipitated product under suction. Recrystallise at least once from industrial spirit to obtain about 2.8 g (44%) of pure 5,5-diphenylhydantoin, m.p. 297-298 °C. [Pg.1153]

The quaternary ammonium cation, tricaprylylmethylammonium, is a well known ion-pairing extracting agent and was used to obtain the ion-pair association complex with 5,5-diphenylhydantoinate anion. The ion-pair complex was embedded in a PVC matrix, containing NPOE as plasticizer. The membrane composition was 7.7% (w/w) electroactive material, 61.5% (w/w) NPOE and 30.8% (w/w) PVC. [Pg.370]
