During the total synthesis of the pentacyclic sesquiterpene dilactone (+)-merrilactone A by S.J. Danishefsky et al., a two-carbon unit was introduced at C9 by a Johnson-Claisen rearrangement. This high yielding transformation was carried out in the presence of catalytic 2,2-dimethyl propanoic acid at 135 °C using mesitylene as the solvent. A mixture of diastereomeric esters were formed, which were later hydrolyzed and subjected to iodolactonization to form the second lactone ring present in merrilactone A. The natural product was synthesized in 20 steps with an overall yield of 10.7%. 

Diethylmethoxyborane is easily prepared by reaction of triethylborane with methanol at room temperature. Small amounts (0.1 mole %) of 2,2-dimethyl-propanoic acid are added to the reaction mixture.2 (Triethylborane reacts irreversibly with 2,2-dimethylpropanoic acid at 0° to give [(2,2-dimethylpro-panoyl)oxy]diethylborane and ethane. With methanol, 2,2-dimethylpropanoic acid and diethylmethoxyborane are formed).

Koch reaction. Carbonium ions react with carbon monoxide (under pressure) to form acyl cations the overall reaction from isobutene gives 2, 2-dimethylpropionic (2, 2-dimethyl propanoic) acid, for example.

Intramolecular Friedel-Crafts acylation has been observed with bonellin dimethyl ester (20).53 The reaction proceeds in contrast to corresponding porphyrins, very smoothly with concentrated sulfuric acid because the propanoic acid side chain at the sp3 center is located above the macrocyclic ring system and therefore can better fulfill the stereoelectronic requirements for the ring-closure reaction. The ring closure is accompanied by racemization in the product 21.

Esters and amides may be sulfinylated. Addition of a mixture of t-butyl acetate and sulfinate ester 19 to a THF-ether solution of magnesium diisopropylamide led to the formation of (R)-(+)-f-butyl p-toluenesulfinylacetate (49) in 90% yield (equation 14)7. t-Butyl propanoate and butanoate also underwent this sulfinylation to give 50 and 51 in yields of 68 and 45%, respectively83. The diastereomeric ratio was 1 1 for 50 and 3 7 for 51. These esters may also be obtained by alkylation of 49. Similarly, treatment of a-lithio-A, A -dimethylacetamide with sulfinate ester 19 gave (R)-( + )-N, Ar-dimethyl-p-toluene-sulfinylacetamide (52) (equation 15)84.
