Chapter 5 Compounds which contain either an ester or an amide link in a single macrocyclic ring. This includes lactones, lactams, carbonates, carbamates, ureas, phosphates, etc. including sulfur, nitrogen and other atoms in the ring. 

Vancomycin, ristocetin A and teicoplanin are produced as fermentation products of Streptomyces orientalis, Nocardia lurida and Actinoplanes teichomyceticus, respectively. All three of these related compounds consist of an aglycone basket made up of fused macrocyclic rings and pendant carbohydrate moieties (Fig. 2-1). The macrocycles contain both ether and peptide linkages. The aglycones of vancomycin and teicoplanin contain two chloro-substituted aromatic rings, while the analogous portion of ristocetin A contains no chloro substituents. 

Intramolecular Friedel-Crafts acylation has been observed with bonellin dimethyl ester (20).53 The reaction proceeds in contrast to corresponding porphyrins, very smoothly with concentrated sulfuric acid because the propanoic acid side chain at the sp3 center is located above the macrocyclic ring system and therefore can better fulfill the stereoelectronic requirements for the ring-closure reaction. The ring closure is accompanied by racemization in the product 21. 

Structural data on ruthenium porphyrins shows that the Ru-N (porphyrin) distance is relatively unaffected by changing the oxidation state, as expected for a metal atom inside a fairly rigid macrocyclic ring (Table 1.11). 

Several toxins produced by marine sponges cause the destabilization of F-actin. They contain a macrocyclic ring and an aliphatic chain, by which they bind to actin protomers. The toxins that include reidispongiolides,... 

One of the first reports related to the preparation of macrocyclic ring systems with boron atoms deals with the transformation of m-difunctionalized borazines into cyclic di-, tetra-, or hexaborazines [16-19]. 

By reaction of 2-alkyl-4,6-dichloro-l,3,5-trimethylborazines (alkyl = methyl, ethyl, i-propyl) with bis(trimethylsilyl)amine the tetrameric borazine ring systems 4-6 are produced (Fig. 2) they can be purified by several successive vacuum sublimations (yields 4-60%). If the borazines carry n-propyl and tert-butyl groups in the 2-position or if methylbis(trimethylsilyl)amine is used to bridge the borazine molecules, the macrocyclic ring formation is inhibited [17, 18]. 

The following structures will show that macrocyclic rings between metal ions or BR2 units and acyloxy or acylamino groups have some relationship with rigid coronands [Ic]. 

A further example of a macrocyclic ring system with boron that is related to the structures of the metallocyclophanes mentioned before is compound 68 that can be prepared via condensation between two bis(borazaphenanthrene) molecules. 

As already mentioned, the macrocyclic ring can be further enhanced if the propylene moiety is substituted by a butylene group. Visible spectral data of a series of related compounds [Cu(L BF2)D]X 132 and the structural study of the derivative with D = H2O show that the seven-membered chelate ring influences the structure and other properties like electronic absorption of the copper(II) complex [193]. 

A a very interesting macrocyclic ring system of the composition Pt(LBp2)2 with L = ethylenediphosphine oxide, Ph(0)HP(CH2)2PH(0)Ph, was also reported a few years ago [204].

The expression bimacrocycle (OT macrocycle or po/ymacrocycle) is used instead of macrobicycle (macrotricycle, etc.) because polymacrocycles can contain further, non-macrocyclic ring systems (e.g. aromatic rings).

The Stille reaction has been successfully applied to a number of macrocyclic ring closures.207 In a synthesis of amphidinolide A, the two major fragments were coupled via a selective Stille reaction, presumably governed by steric factors. After deprotection the ring was closed by coupling the second vinyl stannane group with an allylic acetate.208...
