The purpose of this work was to determine the toxicity to mosquito larvae of insecticide spray residues. That certain insecticides are translocated in plants (4, 5) adds impetus to this study. Fresh orchard fruit sprayed or dusted with preparations containing parathion (0,0-diethyl O-p-nitrophenyl thiophosphate), tetraethyl pyrophosphate (TEPP, HEPP), DDD [2,2-bis(p-chlorophenyl)-l,l-dichloroethane], DDT [2,2-bis(p-chlorophenyl)-l,l,l-trichloroethane], chlorinated camphene, and basic lead arsenate were shipped from California to Yonkers, N. Y., by air express for bioassay. 

The widespread commercial use of DDT [l,l,l-trichloro-2,2-bis-(p-chlorophenyl)-ethane] and the potentialities of the more recently developed parathion (0,0-diethyl O-p-nitrophenyl thiophosphate) have placed emphasis on such investigations concerning these compounds.

The 0,0-diethyl O-p-nitrophenyl thiophosphate used in the preparation of the standard curves was obtained by isolation from a high-purity technical parathion according to the method devised by Edwards and Hall (2). The polarographic method of analysis of parathion as described here has an accuracy of = =1%, and 2 mg. of 0,0-diethyl O-p-nitrophenyl thiophosphate per 100 ml. of solution are apparently a minimum concentration for the sensitivities investigated. However, the polarograph used is equipped with resistors, so that a sensitivity of 0.003 microampere per millimeter may be used. At this sensitivity it would be possible to obtain a sufficient wave height to determine parathion at a concentration of less than 1 p.p.m.

Mills and Hoffmann (1992) investigated ultrasonic degradation of parathion. Parathion (0,0-diethyl O-p-nitrophenyl triphosphate) is a major pesticide used in large quantities worldwide. Organophosphate esters such as parathion have been used as alternatives to DDT and other chlorinated hydrocarbon pesticides however, the organophosphate esters are not rapidly degraded in natural waters. At 20°C and pH 7.4, parathion has a hydrolytic half-life of 108 days and its toxic metabolite, paraoxon, has a similar half-life of 144 days. Ultrasonic irradiation of 25 mL of parathion-saturated, deionized water solution was conducted in a water-jacketed, stainless-steel cell with a Branson 200 sonifier operating at 20 kHz and 75 W/cm2. The temperature of the sonicated solution was kept constant at 30°C. All sonolytic reactions were carried out in air-saturated solution. The concentration of the parathion hydrolysis product p-nitrophenol (PNP) was determined in alkaline solution with a Shimadzu MPS-2000 UV /visible spectrophotometer. 

As Table I illustrates, the chemical classes represented by the pesticides studied include thiophosphates [0,0-diethyl-o-p-nitrophenyl phos-phorothioate], carbamates [1-naphthyI-N-methylcarbamate], dinitrophe-nols [2,4-dinitro-o-sec-butylphenol and 2,4-dinitro-o-cyclohexylphenol], and chlorophenoxy acids [2,4-dichlorophenoxyacetic acid, 2,4,5-trichloro-phenoxyacetic acid, and 2-(2,4,5-trichlorophenoxy)propionic acid]. In addition, a number of molecularly related nitrophenols have been studied to establish the effects of molecular geometry and substituent groups on adsorption of pesticide-type materials.

The metabolic and/or hydrolytic products of parathion encountered as residues in the urine include both diethyl phosphoric acid and diethyl phosphorothioic acid, most probably as their salts (potassium or sodium). Derivatization of these residues with diazomethane would result in the formation of three trialkyl phosphate compounds, namely, 0,0-diethyl O-methyl phosphate (DEMMP), 0,0-diethyl 0-methyl phosphoro-thionate (DEMMTP), and 0,0-diethyl S-methyl phosphorothiolate (DEMMPTh). Earlier (15), it had been shown by combined gas chromatography-mass spectrometry and other analytical data that a later-eluting major product ca. 85%) of the methylation of diethyl phosphorothioic acid formed under the conditions of the analytical method was DEMMPTh, and the minor product formed (ca. 15%) was DEMMTP. Accordingly, all three trialkyl phosphates were observed and confirmed by mass spectrometry in the analysis of the human urine extract. Sufficient internal bond energy differences are associated with the isomeric structures DEMMPTh and DEMMTP that qualitatively and quantitatively dissimilar fragmentation patterns are observed for both isomers as can be seen from the mass spectra of these compounds shown in Figure 4.
