Eig. 5. Several endpoint detection methods were compared for the detection of immuno-polymerase chain reaction (IPCR) amplificate from a direct IPCR (Fig. 3A) of mouse-IgG. Although all IPCR/DNA-detection combinations were able to improve the detection limit of a comparable enzyme-linked immunosorbent assays (ELISA) of approximately 10 amol IgG in a 30-fL sample volume, several differences were observed in actual detection limit, and the linearity of the concentration/signal ratio dependent on the DNA quantification was applied. Best results were obtained for PCR-ELISA (see also Fig. 6) in combination with fluorescence- or chemiluminescence-generating substrates (b, c). With photometric substrates (d) or gel electrophoresis and subsequent spot densitometry (a), a 10-fold decrease in sensitivity was observed. In addition to the more sigmoid curve in gel electrophoresis, an enhanced overall error of 20% compared to 13% in PCR-ELISA was observed for two independent assays. The simple addition of a double-strand sensitive intercalation marker to the PCR-amplificate and measurement in a fluorescence spectrometer further decreased sensitivity (e) and appears therefore to be unsuited for IPCR amplificate quantification. Densitometry analysis to compare between control and treated gels... 

It should be noted that these measurements report the relative abrmdance of each protein in the ceU extracts. Isotope ratios and comparative densitometry do not distinguish changes in expression, regulation, or degradation. Follow-up experiments are required to make these distinctions. 

Samer [137] studied miniemulsion copolymerization in a single CSTR. Two separate feed streams, miniemulsion (or macroemulsion for comparative studies) and initiator were fed at constant rates into the reactor. SLS was used as the surfactant, HD as the costabilizer, and KPS was the initiator. In the miniemulsion configuration (costabilizer included in recipe), the emulsion stream was continuous. Constant volume was provided by an overflow outlet. Salt tracer experiments were used to validate the ideal mixing model assumed for a CSTR. Total monomer conversion was measured via in-hne densitometry, and copolymer composition via offline NMR. 

A. sTF Protein Expression and Analysis. sTF is targeted for secretion by cells, with induced protein observed in the cell-conditioned media. sTF-related proteins from media normally appear as three bands when visualized on 12% polyacrylamide gels the band with the lowest apparent molecular weight is fully processed sTF (Fig. 1). Cells incubated with 5-OHTrp or 7-ATrp express only about 30% media-derived sTF, as measured by densitometry, when compared to the amount obtained with Trp In addition, the relative amount of fully processed protein is less with Trp analogs than with Trp. 

The benefit of using thin-layer chromatography (TLC) for the identification of unknown vitamins and related compounds by comparing Rf values of the unknown compounds with authentic vitamins is beyond doubt. The quantification of the separated vitamins can be performed by the use of modem densitometry. TLC [or high-performance thin-layer chromatography (HPTLC)], as a powerful separation and analytic tool, is used particularly with pharmaceutical preparations and food products. Because amounts of most hydrophilic vitamins are low, or very low, in tissues or body fluids, bioautography or de-rivatization is used before densitometry.
