Column variations

The first packing materials used in a HPLC were beads of organic gel permeation resins used for size separations. These were commercially available resins and no attempt was made to optimize them for high pressure, except to select for a high degree of cross-linkage to prevent crushing. 

A year later silica-based fully porous 35-60-qm diameter beads were slurry packed in a tube and used for separation. This was the same material that had been used for open column or thin-layer chromatography. The only gain over these earlier techniques was in developmental time. Almost immediately, research was begun to optimize the packing in order to improve the separation. 

It was soon found that a large amount of band spreading occurred in this material because of the variety of paths a particle could follow going through the mixture of particle sizes in the packing. Size screening of the packing

One expects this system to have the efficiency of a single column arrangement. However, optimum efficiencies are not realized because of the column-to-column variations and the tendency to use excess feed volumes. 

Many factors can influence the Kovats index which can make it unreliable at times for the characterization of gas chromatographic behavior, although it generally varies less than the relative retention with temperature, flow, and column variation. However, for many it is the preferred method of reporting retention data. 

In the next several years, advances in column technology particularly for the resolution of enantiomers and proteins will continue. This author also expects that sorbent manufacturers will be paying greater attention to minimizing the all too common column-to-column variations.

Even though columns are available that can provide more than 10,000 plates/column or more than 100,000 plates/meter, large column to column variations can be found between manufacturers and from one batch to the next ( ). Experiments were conducted with a permeator composed of 35 silicone rubber capillaries (pressurized internally). Results are presented for the binary systems O2-N2 (air), CO2-N2, CO2-O2, and the multicomponent system C02-CH -N2. Particular attention is given to separation of the C02 CH -N2 mixture in a stripper, conditions for observing composition minima in the enriching section, inherent simulation difficulties in modeling the membrane column, variation of experimental parameters, and local HMU variation along the column. 

A schematic diagram of the Solexol processing of fish oils is shown in figure 7.4. The fish oils are fed to a train of countercurrent extractors where column-to-column variations in temperature as well as temperature variations within each specific column can affect the solvent power of propane in a manner similar to that seen with propane deasphalting.

We use automatic 16-channel pipets (such as the Matrix series from Thermo Scientific) as they allow repeated accurate dispensing of volumes down to 2 pL. Manual alternatives exist, but use of such pipets will increase row-to-row or column-to-column variation and we do not recommend these for screening purposes.
