The thermal conductivity detector (TCD) is a universal detector that is nondestructive, which is a major advantage for preparative work (Dybowski and Kaiser, 2002). However, it is not sensitive enough for many of the analyses discussed later. This detector operates on the principle that a hot body loses heat at a rate dependent on the composition of the material surrounding it (Burtis et ah, 1987). In a TCD, two filaments are heated, one in carrier gas, and the other in the column effluent. The voltages required to maintain the filament at a constant temperature are measured and compared. When compounds elute from the column the voltage of the sample filament is different from that of the filament in carrier gas and is recorded as a peak (Burtis et al., 1987). [Pg.4]

Body fluids, such as serum, contain several different carotenoids in low amounts. The crucial point in the isolation and analysis of these samples is the enrichment factor. Serum samples can be directly analysed with hyphenated extraction-sample enrichment-separation systems, such as on-line SPE-HPLC employing tailored stationary phases [29]. By using special restricted access materials (RAMs) for sample enrichment, the carotenoids are retarded on the pre-column while the protein binding is broken and the macromolecules are eluted. The preparation of artifacts is hindered, as the whole analysis steps take place under conditions of light- and oxygen-exclusion. Micro-injections in micro-flow and nano-flow systems are done with injectors in which the external sample loop is replaced with the internal fixed volume within the injector body. HPLC-on-a-chip systems also build the column into the injector body. The internal path within the injector body is abladed with a laser, packed with micro-packing material, and this serves as the separating media. The injector inlet is connected to the pumping system and the outlet to the detector. Sample is loaded into an internal loop in the load position, then injected onto the chip HPLC by turning the injector. Obviously, in a system like this sample size is very limited and the detector is usually a highly sensitive mass spectrometer. [Pg.116]

HPLC is the technique of choice for determining the purity of moxalactam disodium in raw materials, formulated products, and in body fluids. Moxalactam is determined in a system containing 0.05 to 0.1 M ammonium acetate with about 6 percent methanol present. An ES Industries Chromegabond C18 column or other alternative column with similar retention characteristics is used to determine the purity of the moxalactam sample. The substance may be monitored at 254 nm or, when available, a variable wavelength detector can be operated at 271 nm for assay. The sample is dissolved in water or in 0.1 M ammonium acetate solution. Under conditions of this method, the assay should be completed within 4 hours of sample dissolution. [Pg.324]
