Currently, all five toxic components from concavum are being subjected to column and high performance liquid chromatography. Products of these treatments will be evaluated and characterized using the mouse bioassay and isolated nerve-muscle preparations. 

Ratios were obtained by GC analysis of the crude reaction mixture. Products were separated and isolated by flash chromatography. Products were identified by comparison of their H and 13C NMR spectra with those reported in the literature. Typical isolated yields were 35-40% of 20 and 10-12% of 21...

Calibrations were carried out for the GC/PID or the GC/MS daily. Calibration standards were prepared based on standard reference materials obtained from Supelco Chromatography products. A check standard was analyzed every ten samples to assure calibration and accuracy. A reagent blank was included in each analytic batch of samples. Blanks were made from reagent or make-up water and matrix similar to the sample. A spiked sample was analyzed every twenty samples. This was done by splitting an appropriate sample into two subsamples and adding a known quantity of TCE to one of the split samples. The purpose of a spiked sample is to determine the extent of matrix bias or interference on TCE recovery and sample to sample precision. Accuracy was assessed by analysis of external reference standards (separate from calibration standards) and by percent recoveries of spiked samples. Precision was assessed by means of replicate sample analysis. It is expressed as relative percent difference (RPD) in the case of duplicates or relative standard deviation (RSD) for triplicate (or more) analyses. Recovery was 96% or more for all spiked samples, and RPD/RSD are less than 7% for all samples.

Methanol and an internal standard (acetonitrile or benzene) can be determined with a 2m x 6mm stainless column packed with 30% Triton X-100 on chromosorb W (Gierer et al. 1964, Yang and Goring 1980) or with a 1.8m x 0.32 cm stainless steel column packed with Tenax GC (Lai et al. 1990). The Triton column is available from Supelco Chromatography Products, Bellefonte, PA, while the Tenax column may be obtained from Alltech Associates, Inc., Deerfield, IL. Both columns are operated at 80 °C with an injection port temperature of 150 °C and a detector temperature of 250 °C. A nitrogen flow of 30 ml min-1 is maintained. 

Analysed by gas-liquid chromatography. Products of skeletal rearrangement (see p. 264). 

Determined by gas chromatography, products distilled through a spinning-band column were 94-99% pure.

Pure 3 Is best obtained by chromatography. Product 3 could also be purified by vacuum distillation through a 10-cm Vigreux column, bp 100-103°C (2.2 mm). However, distillation did not efficiently separate 3 from diphenyl disulfide, and bumping was often a serious problem.

Reagents. Unless otherwise indicated, materials were obtained from commercial suppliers and used without further purification. Analytical reagent grade chemicals were used along with deionized water to prepare solutions. Europium (III) oxide, styrene, vinyl benzoate, AIBN, mediyl phosphonic acid and pinacolylmethyl phosphonate were obtained from Aldrich (Aldrich, Milwaukee, WI S3233). Neat analytical standards of the pesticides and insecticides were obtained from Radian (Radian International, Austin, TX 78720). Parathion, methyl-parathion, tfaionazin, and dibutyl chlorendate were obtained as neat standards from Supelco (Supelco Chromatography Products, Bellefonte, PA 16823).
