Cell Capping

Electrochemical cells are assembled in the glove-box. The cell is a 2320-type coin cell (23 mm OD and 2.0 mm thickness) as schematically shown in Fig. 5. The cell includes the electrolyte, the cell cap and can which are stainless steel, a polypropylene gasket used to seal the cell, the two electrodes, the separator between the electrodes, as well as a stainless spacer and a mild steel disc spring which are used to increase the pressure on the electrodes. Once the cell is assembled in the right order, the cell is sealed by a pressure crimper inside the glove-box. [Pg.352]

Normal NMJs have a generally polarized nerve terminal with accumulations of40-50 nm small clear vesicles near the presynap-tic membrane and mitochondria located farther away (Fig. 20.8). In mice, the terminal Schwann cell capping the nerve terminal can be difficult to resolve. The postsynaptic membrane has a series of junctional folds invaginating into the muscle fiber. T-independent immunogens (reviewed by Feldmann, 1974) usually have multiple copies of the same determinant and are, therefore, capable to react with several receptors on the same B cell ( capping ), which activates the cell. At high concentrations, T-independent immunogens may become polyclonal activators of B cells and act as mitogens of B cells. The simultaneous PTA-DTA apparatus is illustrated in Figure 9.20. It consisted of a Stone-Premco DTA cell which was modified by drilling a 0.275-in. opening in the cell cap and furnace chamber to permit sample viewing by the photomultiplier tube. Cell-to-cell contact between MSCs and osteoblasts can enhance communication between cells. Cap junctions, transmembrane channels between neighboring cells that allow communication through the cytoplasm, are present between osteoblasts, osteoclasts, and osteocytes (Civitelli 2008). Connexin 43 is the most abundant gap junction in bone and beyond allowing cells to respond to external stimuli, has been shown to be required for osteoblast differentiation and function (Civitelli 2008, Loewenstein 1981, Schiller et al. 1992). Each gap junction is composed of a hemichannel on each cell that allows for the diffusion of ions, metabolites, and small signaling molecules. Different connexins have varying permeabilities for molecules, allowing selective cell communication. Human osteoblasts were shown to couple to bone marrow stromal cells and allow luciferase dye transfer, a phenomenon that was inhibited by the application of octanol, an inhibitor of gap junction communication (Civitelli et al. 1993). One study proposed cell-to-cell contact to be osteoinductive for MSCs, but the use of cells from different species may have prohibited ideal communication (Kim et al. 2003). [Pg.415]
