Larotaxel (XRP-9881, RPR 109881A) 59 (Sanofi-Aventis) is undergoing Phase III trials in patients with advanced pancreatic cancer who had been previously treated with gemcitabine, as well as in combination with cisplatin to treat locally advanced/metastatic urothelial tract or bladder cancer.124 A Phase III trial for the treatment of advanced breast cancer has been completed. Larotaxel 59129>130 js a semi-synthetic derivative of 10-deacetyl baccatin III with a docetaxel-like side chain that has a low affinity for the P-glycoprotein drug efflux pump, an efflux mechanism that diminishes the effectiveness of the marketed drugs paclitaxel 60 and docetaxel. Importantly, this low affinity should enable larotaxel 59 to be effective in tumours resistant to paclitaxel 60... [Pg.333]

GEMCITABINE ANTICANCER AND IMMUNOMODULATING DRUGS-PACLITAXEL Possibly 1 anti-tumour effect in breast cancer Based on experiments on breast cell lines Avoid co-administration except in clinical trials... [Pg.307]

Fatal cholestatic hver failure occurred in a 45-year-old woman with metastatic breast cancer who was given gemcitabine and carboplatin and pre-existing liver damage. After four courses of gemcitabine -I- carboplatin she developed severe decompensated cholestatic hepatitis (9). Liver biopsy showed marked cholestasis and hepatocellular injury consistent with drug-induced hepatotoxicity. [Pg.1485]

The pharmacokinetics of gemcitabine and doxorubicin did not differ when they were given on the same day, when compared with when they were given alone in patients with breast cancer. Similarly, gemcitabine pharmacokinetics were unchanged by the concurrent use of epirubicin and paclitaxel in patients with breast cancer, and gemcitabine did not alter the interaction between epirubicin and paclitaxel (see Anthracyclines -i-Taxanes , p.612). [Pg.635]

Cardiovascular Cardiac toxicity is rare. Acute myocardial infarction has been reported in patients with underlying cardiac disease [41" ]. Cardiac dysrhythmias are very rare [42" ]. In one case a 67-year-old woman with breast cancer and no cardiac history or risk factors developed a supraventricular tachycardia after an infusion of gemcitabine [43" ]. The dysrhythmia was eventually terminated with oral propranolol and no alternative precipitating cause of the event could be identified. There were no further episodes after withdrawal of the drug. [Pg.735]

Gemcitabine is a 2, 2 -difluoro-2 -deoxycytidine that is used in the treatment of numerous cancers (breast, NSCLC, pancreas, etc.). Its market is wide more than 500 million (U.S.) and 2.5 tons produced in 2001. Synthesis of gemcitabine is... [Pg.280]

Gemcitabine is indicated in the treatment of breast, pancreatic, and nonsmall cell lung cancers. Cytarabine, which can be administered subcutaneously and intrathecally in addition to IV, is used in the treatment of various leukemias. A liposomal formulation of cytarabine is available for the treatment of lymphomatous meningitis. [Pg.1823]
