Blocked amino-terminal

We report a method that facilitates the identification of the blocked amino terminal peptide in a digest of a blocked protein. This method can be applied even if the amino acid sequence and blocking group are unknown. 

The importance of the amino terminal valyl residue for the binding of 2,3-DPG is indicated by the observation that hemoglobins with blocked amino terminal residues, such as the minor human hemoglobin Aic (B77) and one of the hemoglobins of the cat (Tl), and hemoglobins that lack this residue and thus have p chains with only 145 residues, such as the hemoglobin of sheep (B79), do not combine with this organic phosphate.

The hybrid approach enhances the speed and efficiency of primary structure analysis and the range of proteins that can be sequenced. It also circumvents obstacles such as the presence of an amino-terminal blocking group or the lack of a key overlap peptide. Only a few segments of primary strucmre must be determined by Edman analysis.

Copolymers. Copolymers from mixtures of different bisphenols or from mixtures of dichlorosulfone and dichlorobenzophenone have been reported in the patent literature. Bifunctional hydroxyl-terminated polyethersulfone oligomers are prepared readily by the polyetherification reaction simply by providing a suitable excess of the bisphenol. Block copolymers are obtained by reaction of the oligomers with other polymers having end groups capable of reacting with the phenol. Multiblock copolymers of BPA-polysulfone with polysiloxane have been made in this way by reaction with dimethyl amino-terminated polydimethylsiloxane the products are effective impact modifiers for the polyethersulfone (79). Block copolymers with nylon-6 are obtained when chlorine-terminated oligomers, which are prepared by polyetherification with excess dihalosulfone, are used as initiators for polymerization of caprolactam (80). 

The literature data on the preparation of phosphono-dipeptides from 1-aminoalkanephosphonic acids showed 7-7 that the yields of condensation reactions are usually small or moderate. Moreover,the use of bulky N-blocked amino acids drastically decreased the reaction yield. Thus following Martell s method— we wre unsuccesful in the preparation of dipeptides containing N-terminal valine or leucine,while peptides of phenylalanine were obtained in 5—10% yield.Also the active ester method appeared to give small yields of the desired products. Our studies using p-nitrophenyl- and cyanomethyl esters of N-phtaloyl amino acids confirmed these observations. 

Segmented copolymers with polyimides were produced by Keohan [43] and also by Smith [44], who also utilized the same coupling reaction with other amino-terminated engineering thermoplastics (ETP) to produce perfectly alternating block sequences.
