Bleomycin, emulsions

There are fewer reports on other parenteral routes of administration of emulsions such as intramuscular, subcutaneous, and intraperitoneal. V Mer-in-oil and w/o/w emulsions of bleomycin, mytomycin C, and 5-Luorouracil have been administered by these routes, apparently resulting in increased regional lymphatic uptake, hence the term lymphotropic emulsions (Davis et al., 1987). Gasco et al. (1990) developed a w/o emulsion of an LHRH analog measurements of testosterone levels after intramuscular administration to rats suggested that the formulation led to prolonged release of the drug. [Pg.211]

Takahashi et al. [261] have used, with some success, intratumour injection of multiple emulsions of bleomycin and mitomycin in cases of inoperable carcinoma in human patients. Some of the impetus for the increased interest in multiple emulsions has come from the concept of liquid membranes for detoxification. Liquid membranes exist as part of large multiple oil droplets containing many internal dispersed droplets. The system was devised by Exxon Research and Engineering and they have coined the term liquid membrane capsule (LMC) (see Fig. 8.50). For use in vivo the LMCs must survive transit in the gut following oral administration. Asher et al. [262] have studied the mobility of LMCs through the... [Pg.556]

Figure 8.49 Lymph node concentration of bleomycin following intragastric injection of bleomycin or bleomycin-dextran sulphate in the form of (a) aqueous solution or (b) W/O emulsion. [Pg.557]

Injection volume was 50 fi and concentration of injection vehicle was 30 mg ml for bleomycin and l(X)mgml for bleomycin-dextran sulphate complex. Results are expressed as the mean of two animal groups (10 rats). The water-in-oil emulsion comprised 77.8% sesame oil, 1.5% polyoxyethylated castor oil (E = 60) and 5.7% sorbitan sesqui-oleate. From Muranishi et al. [258] with permission. [Pg.557]
