For drug identification and determination of the alkaloid content, DAB 10 describes a TI.C comparison with alkaloid mixtures containing defined ratios of atropine-SO, to scopolamine-HBr (T1-T3). Identification of the drug is then based on the similarity of colour intensity and zone size between the standard solutions and drug extracts. Belladonnae folium (1) the ratio of hyoscyamine (R, 0.25) to scopolamine (R, 0.4) corresponds to that of T1 at about 3 1. Both alkaloids are also pre.senf in the roots and seed.s.

Alkaloids in Belladonnae, Hyoscyami and Stramonii folium (1-3). The tropane alka-loid.s (-)-hyoscyamine (during extraction procedures partly changed into ( ) atropine) and scopolamine as major compounds of the aUcaloidal fraction of Solanaceae drugs respond to Dragendorff reagent with orange, unstable colour. Treatment with NaNO, increa.ses the colour stability of the hyoscyamine zones. 

Scopoliae- (4) and Belladonnae radix (5), which have a similar hyoscyamine to scopolamin content, are characterized by different patterns of blue fluorescent caffeic acid and coumarin derivatives (see Chap. 5 for further information). In Belladonnae (6) and Hyoscyami nigri folium (8), the main zones are rutin (R, 0.4 orange fluorescence) and chlorogenic acid (R,- 0.4.5 blue fluorescence). In Hyoscyami nigri folium, these are... 

Stramonii folium (7) is characterized by five orange fluore.scent quercetin glycosides in the R, range 0.03-0,25. The absence of rutin and chlorogenic add clearly distinguishes the drug from Belladonnae and Hyoscyami folium. Hyoscyami mutici folium (9] has only a very low flavonoid content.
