Automatic application

High-performance thin-layer chromatography (HPTLC) has become widely used and while it follows the same principles as TLC, it makes use of modern technology including automatic application devices and smaller plates, which allow for better sensitivity. 

If magnesium reacts with oxygen, then automatic application of the crisscross rule would lead to the formula Mg202, which is incorrect because the subscripts are not in the lowest whole-number ratio. Planar chromatography, also known as Thin Layer Chromatography (TLC), is a technique related to HPLC but with its own specificity. Manual application of a large number of samples is time-consuming and, obviously, leads to poor reproducibility. Speed and reproducibility are often achieved by using automatic applicators. Such automatic samplers combine precision, accuracy, speed, and versatility. 

A specially modified sampler for Desaga expands the TLC-Applicator AS 30 into a fully automatic application system. The applicator control of the autosampler is performed by a serial interface, and it monitors the selection of sample containers, the delivery of the necessary amount of sample, and the rinsing processes. A second serial interface allows the application parameters to be documented using a printer or laboratory computer system. 

In this chapter, we have outlined the principal motives and methods that pertain to good pharma-covigilance. We have also tried to show how, in particular, risk-benefit analysis must always be on a case-by-case basis, and how it relies, ultimately, on the judgment of those experienced in this field rather than some automatically applicable arithmetic algorithm. Large-scale patient exposures will always trump clinical trials databases for rare types of adverse event, and this continues to be demonstrated by cases such as thalidomide, terfenadine and rofecoxib. 

Jork thinks it is better to use the same capillary for all application operations in an experimental run , provided that it is rinsed at least three times between experiments to prevent errors due to carryover [30]. Here, an experimental nm means the apphca-tion of several samples to one plate for quantitative analysis, as was usual before the use of automatic application equipment. Before purchasing equipment for semiautomatic or fully automatic application, it is advisable to test its performance oneself. Good information can also be obtained from congresses such as the Internationale Symposium fiir Planar-Chromatographie which takes place every 2 years in Interlaken (Switzerland) and exhibitions such as InCom, Diisseldorf (annually), Analytica, Munich (every 2 years) and Achema, Frankfurt (every 3 years).

To get complete coverage of the surface, the gun is moved about and directed to various areas by the sprayman in case the operation is a hand spray one. When arranged as an automatic application, the gun or guns may be mounted on a mechanical manipulation device to produce the desired motion over the surface. The part is often carried on a conveyor to be moved past the spray gun position to further create the desired relative movement of the surface with respect to the spray.

One melting device and pump may serve several dispensing devices. For automatic applications, controls must be available that detect the presence of a substrate, and insure that the adhesive is deposited in the desired location. Both mechanical and optical sensors can be used for these purposes.
