Analysis of serum proteins

Ornstein21 and Davis24 developed the first high-resolution PAGE system for native proteins. Their popular system is still in widespread use. It was designed for the analysis of serum proteins, but works well for a broad range of protein types. The Ornstein-Davis buffers should be the first discontinuous system tried when working with a new, native sample.

Agarose gels with concentrations of 0.7-1% (m/v) are often used in clinical laboratories for analysis of serum proteins and isoenzymes of diagnostic importance such as lactate dehydrogenase and creatine kinase. 

Proteomics have been used to develop biomarkers. This is achieved by comparative analysis of protein expression in healthy and diseased tissues to identify expressed proteins to be used as new markers, by analysis of secreted proteins in cell lines and primary cultures, and by direct serum protein profiling. MALDI seems to be a good technique for direct protein analysis in biological fluids (e.g., the identification of the small proteins-defensin 1, 2 and 3 as related to the anti-HIV-1 activity of CD8 antiviral factor).
